中药及其天然产物通过靶向铁死亡治疗肝癌研究进展

摘要:

肝癌是各种慢性肝脏疾病的终末阶段,发病率及病死率均较高。铁死亡是一种铁依赖性的,脂质过氧化异常累积所导致的调节性细胞死亡方式。近年来大量研究表明铁死亡在肝癌的发生、治疗及耐药方面发挥重要作用。现已有研究表明中药及其天然产物可通过调控谷胱甘肽过氧化物酶4(glutathione peroxidase 4,GPX4)、铁蛋白自噬以及其他一些相关物质水平诱导铁死亡影响肝癌的发生发展。一些天然产物与索拉非尼(sorafenib,SOR)联合应用诱导铁死亡,可增强肝癌细胞对SOR的敏感性、减弱SOR耐药性。因此,本文通过总结多种中药及其有效成分通过诱导铁死亡发挥抗肝癌作用或协同减弱化疗药物的耐药性及作用机制,为肝癌治疗和逆转耐药提供新思路,同时也为开发新型抗肝癌药物提供思路。

Abstract:

Liver cancer is the end stage of various chronic liver diseases, with high incidence and mortality rates. Ferroptosis, an iron-dependent form of regulated cell death mediated by excessive lipid peroxidation, plays an important role in HCC pathogenesis, therapeutic response, and drug resistance mechanisms. ADDIN CNKISM.UserStyleExisting studies have shown that traditional Chinese medicine (TCM) and its natural products can induce ferroptosis and influence the occurrence and progression of liver cancer by regulating glutathione peroxidase 4 (GPX4), ferritinophagy, and other related molecular pathways. Notably, some natural products, when combined with sorafenib (SOR), can enhance the sensitivity of liver cancer cells to SOR and reduce SOR resistance by inducing ferroptosis.Therefore, this review summarizes various TCM and active compounds that exert anti-liver cancer effects by inducing ferroptosis or synergistically mitigating chemotherapy resistance, along with their underlying mechanisms. This provides new insights into liver cancer treatment and drug resistance reversal, while also offering potential strategies for developing novel anti-liver cancer drugs.
