银椴苷的制备和对α淀粉酶的抑制作用

摘要: 银椴苷是从藏药绿萝花中分离得到的一种重要的黄酮类化合物,具有降血糖等活性。本文从藏药绿萝花中制备得到银椴苷,采用以2氯4硝基苯基 αD吡喃麦芽糖苷(CNPG3)为底物的方法进行α淀粉酶活性测试,首次报道了银椴苷对α淀粉酶的抑制作用,并进行了α淀粉酶抑制动力学实验和小鼠淀粉耐量实验。实验结果表明,银椴苷对α淀粉酶具有很强的抑制作用,其半数抑制率(IC50)为12.1 μM。动力学分析表明银椴苷对α淀粉酶的抑制呈非竞争性抑制,抑制常数(Ki)= 9.72 μM。小鼠淀粉耐量实验表明,银椴苷对正常小鼠淀粉负荷糖耐量有明显的改善作用(P<0.05)。

Abstract: The aim of the research was to study the inhibition of tiliroside to αamylase,which was isolated from the flowers of Edgeworthia gardneri.Tiliroside was evaluated for its inhibitory activity against αamylase.The mode of inhibition for αamylase and oral sucrose tolerance test was also investigated.Tiliroside showed significant αamylase inhibitory activity (IC50 = 12.1 μM) in a noncompetitive fashion with a Ki of 9.72 μM.The results obtained from oral sucrose tolerance test revealed that tiliroside is able to inhibit αamylase at the intestinal level with consequent decrease of the postprandial peak in mice.This is the first report on the αamylase inhibition of tiliroside. In vitro and in vivo experiments showed that the inhibition of α amylase is one of the mechanisms of antihyperglycemic activity of E.gardneri.
