新疆千叶蓍中化学成分及其抗氧化和抗菌活性研究

Phytochemical,antioxidant and antibacterial activities of Achillea millefolium L.

摘要: 为探究生长于新疆的千叶蓍(Achillea millefolium L.)中的活性物质基础,本文对采自新疆哈密地区的千叶蓍开展了化学成分分离鉴定及其生物活性研究。利用柱层析色谱、制备型高效液相色谱等方法,从千叶蓍的全草中分离出二十种化合物。通过核磁共振和质谱等波谱学技术以及和文献数据对比,将其分别鉴定为猫眼草黄素(1)、矢车菊黄素(2)、槲皮素(3)、芹菜素(4)、芹黄素-7-O-β-D-葡萄糖苷(5)、(-)-芝麻素(6)、乙氧基阿魏酸(7)、咖啡酸乙酯(8)、香豆酸(9)、对羟基苯丙酸(10)、伞形花内酯(11)、7-甲氧基香豆素(12)、香草酸(13)、4-羟基苯甲酸(14)、二氢猕猴桃内酯(15)、paeoveitol B(16)、2-羟基-2-[(E)-1α,2β,3-三羟基-3-壬烷-5,7-二炔]-4H-吡喃(17)、吲哚-3-乙醛(18)、β-谷甾醇(19)和3β-羟基-5α,8α-环二氧麦角甾-6,22-二烯麦角甾醇过氧化物(20)。其中,化合物20为首次从本植物中分离得到,化合物6、8~10、12~13、15~18为首次从本属植物中分离得到。此外,初步评估了乙醇提取物、乙酸乙酯部位和化合物1~5、8~9、13~14和16的抗氧化及其抗菌活性。化合物3(IC50=9.9±0.94 μM)和8(IC50=21.29±1.65 μM)表现出显著的DPPH自由基清除能力,化合物9、14和16表现出抑制白色念珠菌、大肠杆菌和金黄色葡萄球菌生长的能力。本文首次报道了分布于新疆的千叶蓍中化学成分及其活性,结果表明,酚类化合物如黄酮、苯丙素是该植物中的主要成分。此外,本研究发现千叶蓍中成分表现出一定的抗氧化和抗菌活性,为该植物作的药用价值提供了一定的科学依据。

Abstract: For the aim of reveal the bioactivities and corresponding chemical constituents of Achillea millefolium L.distributed in Xinjiang,China,investigations of isolation,structure elucidation,and bioactivity on A.millefolium collected from Hami region of Xinjiang were carried out.Twenty compounds were isolated from the whole plant of A.millefolium L by various techniques including column chromatography and semi-preparative HPLC.Their structures were identified based on spectroscopic data analysis such as NMR and MASS,together with comparison with previous data,to be centaireidin (1),chrysosplenetin (2),quercetin (3),apigenin (4),apigenin-7-O-β-D-glucoside (5),(-)-sesamin (6),ethoxy-ferulate (7),ethyl caffeate (8),p-couaric acid (9),p-hydroxy benzenepropanoic acid (10),umbelliferone (11),7-methoxy coumarin (12),vanillic acid (13),4-hydroxybenzoic acid (14),dihydroactinidiolide (15),paeoveitol B (16),2-hydroxyl-2-[(E)-1α,2β,3-trihydroxyl-3-nonaene-5,7-diyne]-4H-pyran (17),indole-3-aldehyde (18),β-sitosterol (19),and ergosterol peroxide (20).Among them,compound 20 was isolated from this plant for the first time,and ten compounds (6,8~10,12~13 and 15~18) were firstly discovered from Achillea genus.In addition,antioxidant and antimicrobial activities of the ethanol extract,ethyl acetate fraction,and compounds 1~5,8~9,13~14 and 16 were also evaluated.Compounds 3 (IC50=9.9±0.94 μM) and 8 (IC50=21.29±1.65 μM) showed strong DPPH free radical scavenging capacity.Compounds 9,14 and 16 were found to inhibit the growth of Candida albicans,Escherichia coli and Staphylococcus aureus.This was the first report on the chemical constituent and bioactivity of A.millefolium growing in Xinjiang.Phenolic compounds including flavonoid and phenylpropanoid were found to be the major components in this plant.Moreover,the discovery of antioxidant and anti-microbial activities of compounds in A.millefolium supported the medicinal values of this plant.
