本研究采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱联用技术(UPLC-QE Orbitrap MS/MS)鉴定老虎芋水提物(Alocasiae Cucullatae Rhizoma aqueous extract,ACRAE)及大鼠血清中的化学成分。色谱分离以水(含0.1%甲酸)-乙腈为流动相进行梯度洗脱,流速0.35 mL/min,进样量5 μL。质谱检测采用加热电喷雾电离(HESI)离子源,分别在正、负离子模式下采集质谱信号,扫描范围设置为m/z 100~1 200。通过比对ACRAE、空白血清和含药血清的图谱差异,根据保留时间、相对分子质量,以及一级、二级离子碎片,解析ACRAE经大鼠灌胃给药后血清中的原型成分和代谢产物。ACRAE鉴定得到135个化学成分。在血清中检测到68个入血成分,其中23个为原型成分,45个为代谢产物。通过UPLC-QE Orbitrap MS/MS技术快速分析ACRAE的入血成分,为进一步阐明老虎芋的药效物质基础及作用机制提供了参考。

Abstract:

This study utilized ultra-high-performance liquid chromatography coupled with quadrupole-electrostatic field orbitrap high-resolution tandem mass spectrometry (UPLC-QE Orbitrap MS/MS) to identify the chemical components in Alocasiae Cucullatae Rhizoma aqueous extract (ACRAE) and rat serum. Chromatographic separation was performed using a gradient elution with a mobile phase of water (containing 0.1% formic acid) and acetonitrile at a flow rate of 0.35 mL/min, with an injection volume of 5 μL. Mass spectrometric analysis was carried out with a heated electrospray ionization (HESI) source, and data were acquired in both positive and negative ion modes over a scanning range of m/z 100–1 200. By comparing the chromatographic profiles of ACRAE, blank serum, and drug-containing serum, prototype components and metabolites in the serum after oral administration were identified based on retention times, molecular weights, and both primary and secondary ion fragments. A total of 135 chemical components were characterized in the ACRAE. In serum, 68 blood-absorbed components were detected, including 23 prototype components and 45 metabolites. The rapid profiling of blood-absorbed components in ACRAE via UPLC-QE Orbitrap MS/MS provides critical insights for further exploration of its pharmacodynamic material basis and mechanisms of action.
