甘肃棘豆的化学成分及抗疟活性研究

摘要:

研究高寒地区甘肃棘豆Oxytropis kansuensis Bunge干燥全株的化学成分和抗疟活性。分离和纯化过程采用硅胶、D101大孔树脂、Sephadex LH-20等多种柱色谱方法;根据理化性质及波谱数据鉴定化合物的结构;采用SYBR Green I法,对分离得到的化合物进行抗疟活性筛选。从甘肃棘豆中分离鉴定了10个化合物,分别鉴定为β-谷甾醇亚麻酸酯(1)、羽扇豆酮(2)、β-谷甾醇(3)、9,12,15-十八烷三烯酸甲酯(4)、芹菜素(5)、木犀草素(6)、1-O-反式对香豆酰甘油(7)、1-O-p-coumaroyl-3-O-feruloylglycerol(8)、胡萝卜苷(9)、芹菜素-7-O-β-D-吡喃葡萄糖苷(10)。化合物1作为天然产物首次报道,本文还首次报道了其1H NMR、13C NMR数据。此外,化合物24 ~ 810为首次从甘肃棘豆中分离得到,化合物14 ~ 6810对恶性疟原虫3D7株具有一定抗疟活性。

Abstract:

The chemical constituents and antimalarial activity of Oxytropis kansuensis Bunge from the alpine region were studied. The compounds were separated and purified by a series of chromatographic techniques, including silica gel chromatography, D101 macroporous resin, and Sephadex LH-20 column chromatography. The structures of the compounds were identified on the basis of physicochemical properties and spectral data, and their antimalarial activities were screened using the SYBR Green I method. Ten compounds were isolated and identified from O. kansuensis, namely β-sitosteryl linolenate (1), lupenone (2), β-sitosterol (3), 9,12,15-octadecatrienoic acid, methyl ester (4), apigenin (5), luteolin (6), 1-O-trans-p-coumaroylglycerol (7), 1-O-p-coumaroyl-3-O-feruloylglycerol (8), daucosterol (9), and andcosmosiin (10). Compound 1 was reported for the first time as a natural product, and its NMR data was also reported for the first time. Compounds 2, 4-8 and 10 were isolated from O. kansuensis for the first time, and compounds 1, 4-6, 8 and 10 showed antimalarial activity against Plasmodium falciparum strain 3D7.
