鱼源生物活性肽美白功效及其作用机制的研究

Whitening effect and mechanism of fish derived bioactive peptides

本文以小鼠黑色素瘤细胞B16为对象,研究了不同浓度(5、10和20 g/L)花胶低聚肽(fish swimming bladder oligopeptides,FSBO)对细胞中酪氨酸酶(tyrosinase,TYR)、环磷酸腺苷(cyclic adenosine monophosphate,cAMP)和黑色素含量的影响;比较了10 g/L的FSBO,鳕鱼蛋白肽(cod collagen peptides,CCP)和维生素C(vitamin C,VC)对B16细胞中黑色素生成的影响;同时,以降低B16细胞黑色素含量为目标,筛选了FSBO、CCP和VC的合适添加浓度。研究结果表明,B16细胞中分别添加5、10和20 g/L的FSBO处理24 h,均能显著降低细胞中TYR活性,降低cAMP和黑色素含量;10 g/L的FSBO、VC、CCP对TYR活性和cAMP含量的抑制效果为:VC> CCP>FSBO;在降低黑色素含量方面的效果为:CCP>FSBO=VC;1.25、2.5、10 g/L 3个浓度处理下,随着添加浓度的增加,VC和FSBO添加组黑色素含量逐渐升高,而CCP组黑色素含量逐渐下降。1.25 g/L和2.5 g/L的FSBO与10 g/L CCP减少黑色素含量的效果均较好,与VC相似。研究结果证明FSBO和CCP两种鱼源生物活性肽均可以通过降低TYR活性和cAMP含量来有效减少黑色素含量,具有潜在的美白功效,但二者具有不同的浓度依赖性。该研究结果丰富了生物活性肽护肤活性和作用机制的研究内容,为鱼源生物活性肽用于化妆品原料研究提供理论依据。

Abstract:

This study investigated the effects of fish swimming bladder oligopeptides (FSBO) at varying concentrations (5,10,and 20 g/L) on tyrosinase (TYR) activity,cyclic adenosine monophosphate (cAMP) levels,and melanin content in mouse melanoma cells B16.Additionally,the melanogenesis-inhibitory effects of 10 g/L FSBO,cod collagen peptides (CCP),and vitamin C (VC) were compared.Furthermore,optimal concentrations of FSBO,CCP and VC for reducing melanin content in B16 cells were screened.The results demonstrated that treatment with 5,10,and 20 g/L FSBO for 24 h significantly suppressed TYR activity and reduced both cAMP and melanin levels in B16 cells.At 10 g/L,the inhibitory efficacy on TYR activity and cAMP content followed the order:VC > CCP > FSBO,while melanin reduction efficacy ranked CCP > FSBO = VC.Notably,melanin content progressively increased with higher concentrations (1.25,2.5,10 g/L) of VC and FSBO,but decreased in the CCP -treated group.Both 1.25 g/L and 2.5 g/L FSBO,as well as 10 g/L CCP,exhibited comparable melanin-reducing effects to VC.These findings indicate that FSBO and CCP,two fish-derived bioactive peptides,effectively inhibit melanogenesis by downregulating TYR activity and cAMP levels,albeit with distinct concentration-dependent responses.This study expands the understanding of the skincare potential and mechanisms of bioactive peptides,providing a theoretical foundation for the application of marine peptide derivatives in cosmetic formulations.
