南瓜叶中γ-氨基丁酸的提取及薄层扫描测定

Determination and Extraction of γ-Aminobutyric Acid from Pumpkin (Cucurbita mosehata) Leaves

摘要: 本文在单因素试验基础上,通过正交试验优化了南瓜叶中γ-氨基丁酸的提取条件,并对γ-氨基丁酸的薄层扫描测定方法进行了探索。结果表明,南瓜叶中γ-氨基丁酸的最佳提取条件为:以20%乙醇为溶剂、料液比1:17(w/v)、提取时间1 h,此条件下得到的γ-氨基丁酸的含量为209 mg/100 g。所建立的薄层扫描测定方法在0.125-2.0 mg/mL范围内呈良好线性关系(r2 =0.9991),平均加样回收率(n=4)为99.61%。

Abstract: The extraction conditions and thin layer chromatography scanning (TLCS) analytic method of γ-aminobutyric acid from pumpkin leaves were studied. On the basis of single factor experiments, orthogonal test was investigated. The optimum parameters for extracting γ-aminobutyric acid from pumpkin leaves were determined as follows: the concentration of alcohol was 20%, the ratio of solid to liquid was 1:17(w/v), and the extraction time was 1 h. Under these conditions, the yield of γ- aminobutyric acid was 209 mg/100 g. The calibration curve showed good linearity in the range of 0.125 to 2.0 mg/mL (r2 =0.9991) with the average recovery (n=4) of 99.61%.
