Optimization of Enzymatic Hydrolysis of High Purity Oleuropein from Olea europaea (Olive) Leaf Extract by Response Surface Methodology

摘要: 本文筛选了各种酶系对橄榄苦苷的水解效果,并探讨了pH、温度、时间、酶底物比四个因素对半纤维素酶水解橄榄苦苷效率的影响。在此基础上,采用BBD实验设计方案(Box-Behnken Design)进行三因素三水平响应面分析,考察pH、温度、酶底物比及三者交互作用对油橄榄叶橄榄苦苷降解为羟基酪醇的酶解影响,并对其工艺优化。结果表明,半纤维素酶对橄榄苦苷的降解效果最佳,优于β-葡萄糖苷酶的降解效果。橄榄苦苷酶解制备羟基酪醇的最佳工艺为pH 5、温度55 ℃、酶底物比为1:9.1、时间6 h,得到羟基酪醇含量为(11.31±0.15)%,橄榄苦苷的降解率为98.54%。这将为油橄榄叶橄榄苦苷酶解制备羟基酪醇的工业化生产提供有效借鉴。

Abstract: The selective enzyme was screened from the 10 enzymes with high degradation rate of oleuropein (OE).The process of single factor (pH,temperature,time,the ratio of enzyme quantity and substrate) and a Box-Behnken Design on enzymatic hydrolysis of 81.04% OE olive leaf extract were studied.It was suggested that preparation of hydroxytyrosol from enzymatic hydrolysis of OE in the olive leaves was feasible.The result showed that the performance of hydrolysis treatments was best using hemicellulase as a bio-catalyst,and the high purity of OE in olive extract was beneficial to biotransform OE into hydroxytyrosol (HT).The optimal enzymatic conditions for achieving maximal yield of HT content obtained by the regression were as follows:pH 5,temperature 55 ℃ and the ratio of enzyme quantity and substrate 1:9.1.The experimental result was (11.31±0.15)%,and the degradation rate of OE was 98.54%.The presented results will provide effective foundation for hydroxytyrosol preparation from oleurpein in olive leaf extract.
