吴茱萸总生物碱化学成分及抗单胺氧化酶活性研究

摘要: 对吴茱萸(Evodia rutaecarpa)总生物碱化学成分进行抗单胺氧化酶活性的跟踪分离研究。应用柱色谱、半制备高效液相色谱、制备TLC、凝胶色谱及重结晶等从吴茱萸中共分离纯化得到7个化合物,借助IR、MS、1H NMR、13C NMR等波谱学方法,分别鉴定化合物为:柠檬苦素(1)、Wuchuyuamide(2)、吴茱萸碱(3)、吴茱萸次碱(4)、双吴茱萸碱(5)、Evodol(6)、Evodiagenine(7)。此外,对单体化合物进行了抗单胺氧化酶(monoamine oxidase MAO)活性的测定。研究表明:1、2、4、6和7对单胺氧化酶抑制作用很低,而3和5在终浓度为167 μg/mL时抑制率能达到80%,故进一步测定了其抑制浓度(IC50),分别为55 μg/mL、68 μg/mL。

Abstract: Seven compounds were isolated and purified from Evodia rutaecarpa by silica gel column chromatography,MCI columnchromatography,Sephadex (LH20),HPLC and recrystallization etc.The structures of these compounds were determined based on NMR and ESIMS spectral data,as well as comparing with previous literature data.Seven compounds were determined as limonoid (1),wuchuyuamide (2),evodiamine (3),rutaecarpin (4),dievodiamine (5),evodol (6),Evodiagenine (7).In the next step,all compounds were screened for their antimonoamine oxidase activities.Compounds 3 and 5 showed MAO inhibitory activities with IC50 values of 55 μg/mL、68 μg/mL,respectively,at the concentration of 167 μg/mL.
