不同季节采收的肉苁蓉HPLC指纹图谱比较研究

摘要: 比较不同季节采收的肉苁蓉指纹图谱差异。采用HPLC法,甲醇0.1%甲酸水溶液梯度洗脱,检测波长330 nm,分别建立了秋季、春季采收的肉苁蓉的HPLC指纹图谱,采用LCMS法对共有峰进行定性分析。通过比较发现两个季节采收的肉苁蓉成分差异明显,聚类分析可以完全将两者分开。两个季节采收的肉苁蓉均标定了5个共有峰,但共有峰不完全相同,两者均含有松果菊苷、肉苁蓉苷A、毛蕊花糖苷,春季采收的肉苁蓉中还含有2′乙酰基毛蕊花糖苷和管花苷B,而秋季肉苁蓉中还含有6′乙酰基红景天苷、肉苁蓉苷 B。本文建立的肉苁蓉HPLC指纹图谱分析方法可用于区分肉苁蓉的采收季节,为指导肉苁蓉的规范化种植及确定合理的采收季节提供了实验依据。

Abstract: To compare the differences of HPLC fingerprints of Cistanche deserticola harvested in different seasons.HPLC method was used to establish the fingerprint of Cistanche deserticola harvested in two seasons (spring and autumn).The mobile phase was methanol0.1% formic acid aqueous solution with gradient elution and the chromatogram was measured at the detection wavelength of 330 nm.And the common peaks were preliminarily assigned by LCMS.Clustering analysis showed that there were obvious differences between the two kinds of serial samples.Five common but not exactly same peaks were identified respectively in the two kinds of samples by fingerprint analysis.Echinacoside,cistanoside A and acteoside were found in both groups.However,2′acetylacteoside and tubuloside B were only found in spring samples;2′Acetylsalidroside and cistanoside A were specifically found in autumn samples.The developed HPLC fingerprint analysis method combined with chemometrics can be used to distinguish the harvest season of Cistanche deserticola.This paper can provide the basis for guiding the standardized planting and determining reasonable harvesting seasons of Cistanche deserticola.
