HPLC法测定不同产地三七果梗中6种成分含量

Determination of Six Components in Panax notoginseng Fruit Stems from Different Producing Areas by HPLC

摘要: 为综合评价不同产地来源的三七果梗质量,本文中运用HPLC法对三七果梗中人参皂苷Rb1、Rc、Rb2、Rb3及三七皂苷Fa、Fc 6种单体皂苷含量进行测定。色谱条件为Thermo Scientific Hypersil GOLD C18色谱柱(250 mm×4.6 mm,5 μm),流速:1.0 mL/min,检测波长203 nm,进样量10 μL。结果表明人参皂苷RbRb1、Rc、Rb2、Rb3及三七皂苷Fa、Fc在0.7813~3.0000×102 μg/mL范围内呈良好线性关系;平均加样回收率分别为102.33% (RSD为2.82%)、97.29% (RSD为4.15%)、97.41% (RSD为2.37%)、98.65%(RSD为1.96%)、96.13%(RSD为2.35%)和97.83% (RSD为4.87%)。三七果梗中6种单体皂苷含量为Rb3>Fc>Rb1>Fa>Rc>Rb2,不同产地和生长年限的三七果梗皂苷含量呈现一定的差异性,范围为2.11%~3.84%。本方法操作简便,结果准确,可用于三七果梗中皂苷含量测定,为三七果梗的质量评价和综合开发提供依据。

Abstract: To evaluate the quality of Panax notoginseng Fruit stems in different areas,the HPLC method was established to determine the contents of six components inPanax notoginseng Fruit stems such as ginsenosides Rb1,Rc,Rb2,Rb3 and notoginsenosides Fa,Fc.The conditions were as follows,Thermo Scientific Hypersil GOLD C18 column (250 mm×4.6 mm,5 μm),flow rate:1.0 mL/min,detection wavelength:203 nm,injection volume:10 μL.This method showed a good linearity with the range of 0.7813-3.0000×102 μg/mL.The average recovery of Rb1,Rc,Rb2,Rb3,Fa and Fc was respectively 102.33% (RSD =2.82%),97.29% (RSD =4.15%),97.41% (RSD =2.37%),98.65%(RSD=1.96%),96.13%(RSD=2.35%)and 97.83% (RSD =4.87%).The contents of six components in Panax notoginseng Fruit stems areRb3>Fc>Rb1>Fa>Rc>Rb2,and it showed some differences by area and year,ranging from 2.11% to 3.84%.The method was simple and reliable,and could be used in the quantitative determination of saponins and development in Panax notoginseng fruit stems.
