Optimal Extraction Conditions of Poly-β-hydroxybutyrate (PHB) in Bacillus megaterium by Response Surface Methodology

Abstract: To obtain the optimal extraction conditions of Poly-β-hydroxybutyrate (PHB) in Bacillus megaterium, the effect of chloroform concentration, extraction time and temperature on PHB extraction were studied. On the basis of single factor experiment, Box-Benhnken central composite method was designed for the three factors. In response to PHB extraction value, the extraction conditions were further optimized by response surface method (RSM). The results showed that when chloroform concentration was 69.7%, extraction temperature was 59.7 ℃, extraction time was 3.0 h, the productivity of PHB was 4.437 g/L, reached its maximum.
