Selection and Identification of Baicalin-Producing Endophytic Fungi Isolated from Scutellaria baicalensis Georgi

Abstract: 17 endophytic fungi have been isolated from roots, stems, leaves and flowers of the medicinal plant Scutellaria baicalensis Georgi. The antimicrobial activity of the fungal fermentation broth was detected against 10 kinds of indicator bacteria. At the same time, analysis of the metabolites of these endophytic fungi was done by thin layer chromatography (TLC) and high-performance liquid chromatography (HPLC). The results of the antimicrobial test showed that the fermentation broths of the 12 strains of endophytic fungi displayed growth inhibition on at least one test bacterium, some of them (G2, J4, J5) showed a broader antimicrobial spectrum. TLC and HPLC analysis showed that 2 strains of fungi from the stem and flower of Scutellaria baicalensis Georgi produced bioactive component baicalin in the artificial culture medium. Colony morphology of the fungal culture, microscopic observation and ITS sequence analysis together indicated that strains J1 and H3 were classified to Penicillium sp.
