姜黄素与5-FU联用对结肠癌SW480细胞的生长抑制与凋亡诱导作用的研究

Effects of Curcumin Combined with 5-Fluorouracil on Growth Inhibition and Apoptosis Induction in Colon Cancer Cell Line SW480

摘要: 本文应用MTT检测法、基于EB/AO双染的荧光显微观察法和FITC/PI双染的流式细胞检测技术,从细胞生长、细胞凋亡和细胞周期等方面研究了姜黄素与5-FU联用对结肠癌SW480细胞的效应。结果显示,姜黄素(25 μM)分别与低剂量(2.4 μM)、中剂量(4.8 μM)5-FU联合作用能有效地抑制SW480细胞的生长,诱导细胞凋亡并将细胞周期阻滞在S期。姜黄素与低剂量5-FU联用组的效果好于中剂量(4.8 μM)5-FU单用组(P<0.05),而姜黄素与中剂量5-FU联用组的效果好于高剂量(9.6 μM)5FU单用组(P<0.05),提示姜黄素能增强5-FU抗结肠癌SW480细胞的作用或对5-FU有协同效应。本研究为临床上应用姜黄素作为5-FU治疗结肠癌的辅助用药,以减少5-FU的使用剂量,从而降低其毒副作用提供了初步的实验依据。

Abstract: In this study,by using MTT assay,EB/AO double staining fluorescence microscopy and FITC/PI double staining flow cytometry,we investigated the combined effects of curcumin and 5-FU on growth,apoptosis and proliferation in human colon cancer cell line SW480.The results showed that curcumin (25 μM),combining with low dose (2.4 μM) and middle dose (4.8 μM) of 5-FU,can effectively inhibit growth,induce apoptosis and arrest cell cycle in S phase of SW480 cells.The combined inhibitory effect of curcumin with low-dose 5-FU group was stronger than that of the middle-dose (4.8 μM) 5-FU group (P<0.05);Similarly,the combination of curcumin with middle-dose 5-FU showed better inhibitory effects when comparing to the high-dose (9.6 mol/L) 5-FU group(P<0.05),suggesting that curcumin can enhance the antitumor effect of 5-FU against SW480 cells,or have the synergic effect to 5-FU.Our study provided a preliminary experimental basis for using curcumin as an adjuvant to 5-FU with the benefit of reducing its toxicity in the clinical treatment of colon cancer.
