银杏叶聚戊烯醇亚急性毒理学研究

Evaluation of Subacute Toxicity of Polyprenols from Ginkgo biloaba L.

摘要: 本文研究了银杏叶聚戊烯醇(GBP)的亚急性毒理实验,为GBP开发为保健食品或医药品提供基础。使用80只SD大鼠,雌雄各半,GBP药物随机分为417、833 mg/kg和1250 mg/kg三个剂量组(相当于推荐剂量的50、100和150倍)和阴性对照组,将GBP掺入基础饲料中连续喂养30 d后,作血常规和生化指标测定,同时取出肝、肾、脾、睾丸称重,并对肝、肾、脾、胃肠、睾丸、卵巢作组织病理学检查。结果表明,GBP各剂量组给予大鼠喂养30 d,动物活性和生长正常;各剂量组体重、30 d食物利用率、脏体比与对照组比较无显著性差异(P>0.05);对雌雄大鼠红细胞计数(RBC)、白细胞计数(WBC)、血红蛋白(HGB)、白细胞计数分类中嗜中性粒细胞(NE)、淋巴细胞(LY)、单核细胞(MO)、嗜酸性粒细胞(EO),嗜碱性粒细胞(BA)等血常规指标没有影响(P>0.05),对血清中天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、尿素(BUN)、肌酐、胆固醇、甘油三脂、血糖、白蛋白及总蛋白等各项生化指标没有产生明显差异(P>0.05),对受检脏器作组织病理学检查未见特异性病变。大鼠连续喂养30 d摄入1250 mg/kg剂量的GBP,未见该受试物明显的毒理效应,对机体未见不良影响。因此,可认为GBP服用安全,无毒副作用。

Abstract: The aim of this study was to investigate the subacute toxicity of polyprenols from G.biloaba L.(GBP).80 rats (female and male,each half) were randomly divided into 4 different groups,including GBP 417,833 mg/kg,and 1250 mg/kg (equivalent to 50,100 and 150 times of the recommended dose) groups and negative control group.When rats were continuously feeding in 30 days with GBP mixed into basal diet,routine test and biochemical indexes of blood were determined.The quantity of liver,kidney,spleen and testis were weighed,and histopathological examination were performed on liver,kidney,spleen,stomach,ovary and testis.During the period of 30 d’s feeding,the results showed that all rats performed normal at the end of the experiment,and the difference between different groups on the body weight,increasing weight,food intake,food utilization rate and organ/body weight ratio of rats were not statistically significant (P>0.05).The difference of routine indexes of RBC,WBC,HGB,NE,LY,MO,EO,BA as well as biochemical indexes of alanine aminotransferase,aspartate aminotransferase,urea,creatinine,blood glucose,total cholesterol,triglyceride,albumin and total protein,were also not statistically significant (P>0.05).Histopathological examination showed no specific lesions for organs.No apparent toxicity and no adverse effects to rats were observed by feeding 1250 mg/kg of GBPfor continuous 30 d.Therefore,GBP can be considered safe and nontoxic.
