槲皮素减轻紫杉醇致神经病理性大鼠疼痛的作用与机制

Effect and mechanism of quercetin on pain relief induced by paclitaxel in neuropathic rats

摘要: 为探究槲皮素对紫杉醇致神经病理学疼痛大鼠的改善作用。实验采用给予雄性SD大鼠腹腔注射4 mg/kg紫杉醇一周来诱导神经痛模型,并给予30、50、100 mg/kg槲皮素连续灌胃7天进行治疗,通过Von Frey测试评价机械性异常疼痛,检测巨噬细胞在大鼠背根神经节(DRG)上的迁移效应,将DRG细胞与紫杉醇和槲皮素共同孵育后观察神经突起的生长,通过Western blotting评价Wnt/β-catenin信号通路变化,结果显示槲皮素能够以剂量依赖的方式的显著升高神经痛大鼠的缩抓阈值,降低巨噬细胞的迁移及由紫杉醇处理引起的大鼠脊髓中炎症因子TNF-α及IL-1β的水平的升高,并可以剂量依赖的方式抑制由紫杉醇引起的大鼠背部神经节神经元突起退化和Wnt信号通路的关键分子Wnt3α和β-catenin蛋白的水平,提示槲皮素可能通过调控Wnt/β-catenin信号通路,影响脊髓中炎症因子的水平并降低炎症细胞的迁移对紫杉醇致神经病理学疼痛大鼠的发挥改善效应。

Abstract: This study investigated the effect of quercetin on paclitaxel-induced neuropathic pain in rats.A model of neuralgia was induced by intraperitoneal injection of 4 mg/kg paclitaxel for 30 weeks,and 30,50 and 100 mg/kg quercetin was administered continuously.After 7 days of gavage treatment,the mechanical abnormal pain was evaluated by Von Frey test,and the migration effect of macrophages on rat dorsal root ganglion (DRG) was detected.The DRG cells were incubated with paclitaxel and quercetin to observe the nerve.The growth of the neurites was evaluated by Western blotting.The results showed that quercetin could significantly increase the shrinkage threshold of rats with neuralgia in a dose-dependent manner,and reduce the migration of macrophages.Increased levels of inflammatory cytokines TNF-α and IL-1β in rat spinal cord induced by paclitaxel,and a dose-dependent manner to inhibit paclitaxel-induced neuronal neurite outgrowth and key molecules of Wnt signaling pathway in rat dorsal ganglion neurons The levels of Wnt3α and β-catenin proteins suggest that quercetin may affect the level of inflammatory factors in the spinal cord and reduce inflammation by regulating the Wnt/β-catenin signaling pathway.Taxol induced cell migration neuropathology of pain in rats exert improvement effects.
