叉枝鸦葱总黄酮清除DPPH自由基活性研究

DPPH Free Radical Scavenging Efficiency of Total Flavonoids from Scorzonera divaricata

摘要: 以叉枝鸦葱为研究对象,用微波辅助提取法提取总黄酮,采用DPPH法研究叉枝鸦葱总黄酮对自由基的消除作用。同时,采用Graphpad Prism 5.0软件进行动力学模型拟合。结果表明:叉枝鸦葱总黄酮对DPPH自由基具的清除作用大于芦丁和Vc,清除率为41.72%~87.72%,IC50为5.6 μg/mL。动力学模型拟合后所得方程Y=(Y0-NS)*exp(-K*X) + NS,充分反映了叉枝鸦葱总黄酮对DPPH自由基的实际消除效应,在样品浓度为20 μg/mL时效应关系最佳。

Abstract: To investigate the antioxidant activity of total flavonoids in Scorzonera divaricata, microwave-assisted extraction method was used to extract total flavonoids. Antioxidant activity was measured by DPPH free radial scavenging assay.Meanwhile,the dynamic model of response curves was simulated by Graphpad Prism 5.0 software.The results showed that the total flavonoids had higher scavenging activity on DPPH free radial than Vc and rutin,clearance rate was 41.72%-87.72%, IC50 was 5.6 μg/mL.A kinetics equation was established:Y=(Y0-NS)*exp(-K*X) + NS,which very well reflected the efficiency of free radical scavenging.The appropriate concentration of the sample was 20 μg/mL.
