山茱萸对肾间质纤维化的保护作用及机制研究

Protective Effect of Cornus officinalis on Renal Interstitial Fibrosis and Its Mechanism

摘要: 为评价山茱萸对肾间质纤维化的保护作用及其作用机制。36只5周龄体重相近的雄性SD大鼠随机分为假手术组(S组)、模型组(U组)、山茱萸(300 mg/Kg)干预组(D组)。分别于术后第7 d、第14 d处死三组中的6只SD大鼠。观察肾组织病理改变和肾间质纤维化程度、YKL-40、β-catenin、Wnt4蛋白表达。结果显示D组各时间点肾小管间质损伤指数、肾间质纤维化指数、YKL-40、β-catenin均低于同期U组,D14组高于D7组(P<0.05﹚。而D组Wnt4和β-catenin、YKL-40蛋白表达量显著降低,与U组相比,差异具有统计学意义(P<0.05)。表明山茱萸对肾脏具有一定的保护作用。

Abstract: The purpose of this study was to evaluate the protective effect of Cornus officinalis on renal interstitial fibrosis and its mechanism.Thirty-six 5-week-old male SD rats with similar body weight were randomly divided into sham group (S group),model group (U group) and C.officinalis (300 mg/Kg) in the intervention group (group D).Six SD rats from each of the three groups were sacrificed respectively at the first 7 days and the first 14 days.Pathological changes of renal interstitial renal fibrosis and YKL-40,β-catenin,Wnt4 protein were observed.The results showed that among D group at each time point tubulointerstitial damage index,renal interstitial fibrosis index,YKL-40,β-catenin were lower than that in U group at the same period,and the indicators in D14 group were all higher than that in D7 group (P<0.05).While Wnt4 and β-catenin,YKL-40 protein expression in group D was significantly decreased compared with the U group (P<0.05).It showed that C.officinalis had a protective effect on kidney.
