无梗五加果实提取物抗肿瘤作用研究

Antitumor Activity of Acanthopanax sessiliflorus (Ruqr.et Maxim) Seem.Fruit Extract

摘要: 研究无梗五加果实提取物对荷瘤小鼠肿瘤生长的抑制作用,及其对体重、胸腺指数、脾脏指数、细胞因子水平等的影响。将H22肝癌细胞悬液接种于小鼠右前肢腋窝皮下(0.2 mL/只)建立小鼠肝癌H22肿瘤模型,然后将90只昆明小鼠随机均分为正常组(等量蒸馏水)、模型组(等量蒸馏水)、阳性组(环磷酰胺0.03 g/kg)、无梗五加果实精多糖高低剂量组(0.80、0.20 g/kg)、无梗五加果实粗多糖高低剂量组(1.04、0.26 g/kg)以及无梗五加果实醇提物高低剂量组(0.92、0.23 g/mL),每天灌胃给药1次,连续给药10 d。采用ELISA法检测荷瘤小鼠血清中白细胞介素2(IL2)、白细胞介素6(IL6)、肿瘤坏死因子α(TNFα)的含量。并称定瘤重计算各给药组的抑瘤率,称定胸腺和脾脏的重量计算胸腺指数和脾脏指数。实验结果显示,与正常组比较,模型组小鼠血清中细胞因子IL2、IL6、TNFα的水平显著降低,瘤重增加,胸腺和脾脏指数显著下降,差异有统计学意义(P<0.05);与模型组比较,无梗五加醇提物高低剂量组、粗多糖高剂量组、精多糖高低剂量组对小鼠移植性实体瘤H22均有抑制作用,抑瘤率分别为 53.27%、52.87%、51.98%、31.64%、38.14%,其中以无梗五加果实醇提物抑瘤作用最显著。并且无梗五加果实各给药组(除粗多糖低剂量组)均能提高荷瘤小鼠的胸腺指数、脾脏指数以及血清中IL2、IL6、TNFα的水平,差异有统计学意义(P<0.05)。无梗五加果实醇提物和多糖对小鼠H22移植性实体瘤均有较好的抑制作用。

Abstract: To study the inhibitory effect of Acanthopanax sessiliflorus (Ruqr.et Maxim) Seem.fruit extract on tumor growth and the levels of cytokines,mice weight,thymus and spleen index in tumorbearing mice.H22 tumorbearing mice model was induced by H22 cell vaccination in normal mouse forelimb right axillary subcutaneous (0.2 mL/rat),then 90 KM mice were randomly divided into normal group (constant volume of distilled water),model group (constant volume of distilled water),cyclophosphamide group (0.03 g/kg),and the refined A.sessiliflorus polysaccharides highdose and lowdose groups (0.80,0.20 g/kg),the crude A.sessiliflorus polysaccharides highdose and lowdose groups (1.04,026 g/kg),the A.sessiliflorus ethanol extract highdose and lowdose groups (0.92,0.23 g/mL).They were given medicine intragastrically once a day for consecutive 10 days.The levels of IL2,IL6 and TNFα in tumorbearing mice were investigated by ELISA assay,the tumor weight and the tumor inhibition rate were calculated and the weight of thymus and spleen were weighted and the indexes of them were calculated.Compared with normal group,the contents of IL2,IL6,TNFα thymus and spleen index decrased significantly in model group.While tumor weight increased,there was statistical significance (P<0.05);Compared with model group,the A.sessiliflorus ethanol extracts highdose and lowdose groups,the crude A.sessiliflorus polysaccharides highdose groups,the refined A.sessiliflorus polysaccharides highdose and lowdose groups had marked inhibitory effects on tumor growth in vivo,the inhibition rates of A.sessiliflorus fruit extract on H22 was 53.27%,52.87%,51.98%,31.64%,38.14%,especially the ethanol extracts from A.sessiliflorus was the strongest one for anticancer activity.It could increase spleen index and thymus gland index and enhance the expression levels of IL2,IL6 and TNFα in tumorbearing mice,there was statistical significance (P<0.05).The ethanol extracts from A.sessiliflorus and polysaccharides can inhibit tumor growth of H22 tumorbearing mice.
