樟树抗炎活性及作用机制研究

Anti-inflammatory Activity of Cinnamomum camphora Fractions and Its Anti-inflammatory Mechanism

摘要: 以樟树提取香料后的残渣为研究对象,探讨其对脂多糖(LPS)刺激RAW 264.7细胞的体外抗炎作用及其机制。采用95%乙醇提取,然后进行萃取分为正己烷、二氯甲烷、乙酸乙酯、正丁醇和水5部分,采用噻唑蓝(MTT)法检测不同萃取相对RAW 264.7细胞活性的影响;采用Griess法检测各萃取相 对LPS刺激RAW 264.7细胞的一氧化氮(NO)释放量;采用酶联免疫吸附法(ELISA)检测细胞上清液中PGE2和TNF-α的分泌量;采用反转录PCR(RT-PCR)和实时定量PCR(real time RT-PCR)检测环氧合酶-2 (COX-2)、诱导型一氧化氮合酶(iNOS)和肿瘤坏死因子-α(TNF-α)mRNA的表达。樟树乙酸乙酯萃取相能明显抑制LPS诱导的RAW264.7巨噬细胞NO、PGE2、和TNF-α的分泌而且无细胞毒性,从转录水平不同程度地抑制COX-2、iNOS和TNF-α的表达。证实了樟树乙酸乙酯萃取相具有抗炎作用,其作用机制与在转录水平上抑制COX-2、iNOS和TNF-α有关。

Abstract: To observe the anti-inflammatory effect of the residue left after extracting spices from Cinnamomum camphora on LPS-induced RAW 264.7 macrophages.Hexane,dichloromethane(CH2Cl2),ethyl acetate(EtOAc),n-butanol (n-BuOH) and water (H2O) fractions of 95% alcohol aqueous extracts of C.camphora were obtained.Then,the effects of five fractions of different concentrations on RAW264.7 cell viability were determined with the MTT method.NO concentration was measured with Griess method.The enzyme linked immunosorbent assay (ELISA) was used to detect the secretion of PGE2 and TNF-α.The reverse transeriptase-polymerase chain reaction (RT-PCR) and real time RT-PCR were used to determine the expression of COX-2、iNOS and TNF-α mRNA.Pre-treatment with EtOAc fraction of C.camphor showed no cytotoxicity can significantly inhibited the LPS-stimulated secretion of NO,PGE2,and TNF-α into the culture medium,as well as the mRNA levels of inducible nitric oxide synthase (iNOS),cyclooxygenase-2 (COX-2),and TNF-α.Overall,our data suggest that EtOAc fraction of C.camphor possess significant anti-inflammatory properties by down-regulation of COX-2、iNOS and TNF-α expression from transcriptional levels.EtOAc fraction of C.camphor may exert anti-inflammatory effect on RAW264.7 cells stimulated by LPS.
