海参皂苷Des-echinoside A通过NF-κB信号通路抑制人肺巨细胞癌95D细胞转移

Triterpene Glycoside Des-echinoside A Inhibits Human Lung Cancer Cell Line 95D Metastasis via NF-κB Signaling Pathway

摘要: 采用高速逆流色谱法从革皮氏海参中分离纯化得到皂苷单体Des-echinoside A,研究了Des-echinoside A对高转移人肺巨细胞癌95D细胞增殖和转移的影响及其分子机制。结果显示:Des-echinoside A能显著降低95D细胞增殖(P<0.01)、迁移(P<0.01)、侵袭(P<0.01)以及粘附能力(P<0.01);还能显著抑制鸡胚尿囊膜(chick embryo chorioallantoic membrane,CAM)上的血管新生。Des-echinoside A能显著降低uPA、MMP-2/-9、VEGF和NF-κB mRNA的表达(P<0.05,P<0.01),显著上调TIMP-1/-2 mRNA的表达(P<0.01),提示Des-echinoside A可能通过调控NF-κB信号通路抑制95D细胞转移。

Abstract: A triterpene glycoside Des-echinoside A was separated by high speed counter current chromatography from sea cucumber Pearsonothuria graeffei. The effects and molecular mechanism of Des-echinoside A on tumor metastasis in high-metastatic (95D) human lung cancer cells were evaluated.The results showed that Des-echinoside A could inhibit 95D cell proliferation (P<0.01),migration (P<0.01),invasion (P<0.01),adhesion(P<0.01) and decrease the blood vessel growth on the chick embryo chorioallantoic membrane(CAM) remarkably.Des-echinoside A could also reduce the mRNA expression of uPA,MMP-2/-9,VEGF,NF-κB (P<0.05,P<0.01),and promote the mRNA expression of TIMP-1/-2(P<0.01).It is suggested that Des-echinoside A could prevent the metastasis through specific inhibition of NF-κB signaling pathway.
