太子参参须皂苷的免疫佐剂作用研究

摘要:

本实验旨在研究太子参参须皂苷(Radix Pseudostellariae fibrous roots saponins,RPFRS)对免疫模式抗原Ovalbumin(OVA)小鼠的免疫佐剂作用。将60只雌性ICR小鼠随机分成对照空白组(CK),单独免疫OVA组(OVA),OVA+RPFRP低、中、高(50、100、200 mg/kg)剂量组(D、Z、G)。在实验的第1~5天和第16~20天,除CK组和OVA组小鼠灌胃双蒸水之外,其他组分别灌胃对应剂量的RPFRS。第6天和第21天,除CK组0.2 mL/只皮下注射PBS外,其余各组小鼠注射配OVA溶液0.2 mL/只,共免疫2次。第36天采样检测相关指标。结果表明:与OVA组相比,RPFRS组对T细胞和OVA的刺激指数均差异显著(P<0.05),D组B细胞刺激指数显著提高(P<0.05),D组和G组IL-2、IL-4含量显著升高(P<0.05),G组IFN-γ含量显著升高(P<0.05),各剂量RPFRS组OVA-IgG和OVA-IgG1含量显著提高(P<0.05),D组和G组OVA-IgG2a含量显著提高(P<0.05),D组和Z组的OVA-IgG2b含量显著提高(P<0.05),RPFRS组IL-2 mRNA、IL-4 mRNA和IFN-γ mRNA相对表达量提高(P<0.05),G组IL-10 mRNA相对表达量提高(P<0.05),RPFRS组CD3+、CD8+ T细胞数和CD4+/CD8+比值显著提高(P<0.05),对CD4+ T细胞的提升作用以D、Z组最为明显(P<0.05)。上述结果表明RPFRS能够增强小鼠免疫功能,提高OVA免疫原性,加强小鼠对OVA免疫应答能力,从而起到一定佐剂作用。

Abstract:

To study the adjuvant effect of Radix Pseudostellariae fibrous roots Saponin (RPFRS) on mice immunized with Ovalbumin (OVA).60 ICR female mice were randomized into control group (CK group),individual immunization OVA group (OVA group),RPFRS low,medium and high dose groups (50,100,200 mg/kg) (D,Z,G groups).On day 1st-5th,16th-20th of the experiment,the other groups were given the corresponding dose of RPFRS except CK and OVA group.On the 6th and 21st day,except for the CK group,0.2 mL of PBS was injected subcutaneously,and 0.2 mL of OVA solution dissolved in PBS was injected into the other groups,that is,the injection weight of OVA in the other groups was 100 μg,twice in total.During the period,each group was free to eat and drink.On the 36th day,related indicators were tested.The results showed that compared with OVA group,the RPFRS groups significantly raised the stimulation index of T cells and OVA(P<0.05).The stimulation index of B cells in D group was significantly increased (P<0.05),the contents of IL-2 and IL-4 in group D and G were significantly increased (P<0.05),and the contents of IFN-γ in group G was significantly increased (P<0.05).The contents of OVA-IgG and OVA-IgG1 in RPFRS groups were significantly increased (P<0.05),the contents of OVA-IgG2a in group D and G were significantly increased (P<0.05),the contents of OVA-IgG2b in group D and Z were significantly increased (P<0.05).The relative expression of IL-2 mRNA,IL-4 mRNA and IFN-γ mRNA in RPFRS groups were significantly increased (P<0.05),and the relative expression of IL-10 mRNA in G group were significantly increased (P<0.05).The number of CD3+, CD8+ T cells and the ratio of CD4+/CD8+ in RPFRS groups were significantly increased (P<0.05),and the promoting effect on CD4+ T cells was the most obvious in group D and Z (P<0.05). In summary,RPFRS can enhance the immune function of mice,improve the immunogenicity of OVA and enhance the immune response ability of mice to OVA,thus playing a certain role as an adjuvant.
