密花香薷总黄酮体外抗氧化及抗肿瘤活性研究

In vitro Antioxidant Activity and Anti-tumor Activity of Total Flavonoids from Elsholtzia densa Benth.

摘要: 通过超声波法提取、D101大孔吸附树脂纯化密花香薷总黄酮,体外抗氧化试验和MTT(四甲基偶氮唑盐)法评价其抗氧化和抗肿瘤活性。结果表明,总黄酮对羟自由基(·OH)、DPPH自由基(DPPH·)和超氧阴离子自由基(O·2)均具有清除能力,其中对O·2的清除能力优于VC;MTT实验结果显示总黄酮对4株细胞均有不同程度的抑制效应,并呈现时间和剂量依赖性(P<0.05)。其中对人宫颈癌Hela细胞48 h抑制效应最明显,IC50值为0.5 mg/mL;形态观察显示,在总黄酮作用下,SMMC-7721和Hela细胞逐渐变圆,粘附性降低,细胞表面皱缩,产生细胞碎片,显示出明显的细胞毒性,AO/EB染色观察发现随着总黄酮处理浓度的增加,凋亡细胞的数量也在逐渐增加。本研究结果表明密花香薷总黄酮具有很好的抗氧化活性,可作为一种天然抗氧化剂,且总黄酮对肿瘤细胞生长有一定的抑制效应,可作为抗肿瘤药物进一步研究。

Abstract: The total flavonoids from Elsholtzia densa Benth.were extracted by ultrasonic extraction method and purified with D101 macroporous adsorption resin. In vitro antioxidant test and MTT assay were used to evaluate its antioxidant and anti-tumor activity.The results showed the total flavonoids had good scavenging ability on hydroxyl radical (·OH),DPPH free radical (DPPH·) and super oxide anion radical (O·2).Its scavenging capacity for O·2 was better than that of VC.MTT assay results showed that the total flavonoids from E.densa significantly inhibited the growth of four cell lines in a dose and time-dependent manner (P<0.05).At 48 h treatment,the inhibitory effect of flavonoid was better on human cervical carcinoma Hela cells (IC50 0.5 mg/mL) than other cells.Morphological observation revealed that SMMC-7721 and Hela cell lines showed cytotoxicity including cell wrinkle,reduction of cellular cohesiveness and breakage of cell membranes after treatment with the E.densa total flavonoids.AO/EB staining showed that the number of apoptotic cells was gradually increased with the increasing of the concentration of total flavonoids.The results suggested that E.densa total flavonoids had excellent antioxidant activity and can be used as a natural antioxidant.It also had inhibitory effect on growth of tumor cells,and can be used as a potential anticancer drug for further study.
