白术、桂枝挥发油β-环糊精包合工艺研究及其包合物评价

Preparation and Evaluation of Rhizoma Atractylodis macrocephalae and Ramulus cinnamomi Oil β-cyclodextrin Inclusion Complex

摘要: 本实验研究以β-环糊精(β-CD)为材料,筛选包合白术、桂枝混合挥发油的最佳工艺并对其进行表征。采用正交试验分别对搅拌法、研磨法、超声法三种方法进行比较,以混合挥发油收得率、包合率、含油率为指标,优选出最佳包合方法包合挥发油的最优工艺;通过紫外分光光度法(UV)、薄层色谱法(TLC)、差示热分析法(DSC)以及显微成像对包合物进行物相鉴别分析。最终优选的最佳包合方法为饱和水溶液法,工艺条件为:挥发油与β-CD投料比为1:8,包合时间为3 h,包合温度为40 ℃;经UV、TLC、DSC以及显微成像进行表征后,分析结果均显示挥发油与β-CD确已形成包合物。且该工艺制备的环糊精包合物的包合率高,工艺简便、可靠,为将葶苈生脉方开发为现代中药制剂提供实验依据。

Abstract: The aim of this study was to optimize the inclusion technology of volatile oil with β-CD and to use appropriate methods for identification.Three methods including saturated water solution,grinding and ultrasonic were investigated to prepare inclusion compound.And the method was established based on the inclusion rate of volatile oils,yield of inclusion compound and oil content.After that,the formulation and processing parameters were optimized by L9(34) orthogonal design.The formations of inclusion complex were validated by UV,TLC,DSC and microscopic imaging.The optimal preparation conditions for inclusion were established as follows:the method of saturated solution stirring was the best and volatile oil: β-CD was 1:8,the inclusion temperature was 40 ℃ and the time was for 3 hours and the inclusion temperature was at 40 ℃.Analysis results showed that the inclusion technique was successful.The process was simple and it can improve the stability and solubility of volatile oil effectively.Furthermore the method can be used for industrial production of Tingli Shengmai decoction.
