苦豆碱对小鼠的急性毒性研究

Acute Toxicity of Aloperine on Mice

摘要: 本文拟探讨苦豆碱对小鼠的急性毒性,并明确其半数致死量(LD50)以及毒性靶器官。采用腹腔注射给药方式,给予600.00、480.00、384.00、307.20、245.76 mg/kg的苦豆碱,实验后观察7 d,记录体重变化及自主活动情况,检测苦豆碱对小鼠的急性毒性作用,用加权回归几率法(Bliss)计算LD50,并对死亡小鼠进行主要脏器的病理学检查。结果显示,苦豆碱的小鼠LD50为395.62 mg/kg,95%可信限为305.23~535.04 mg/kg,LD50为143.00 mg/kg,LD95为1094.47 mg/kg。7 d内小鼠体重有增长趋势。病理结果显示,苦豆碱对小鼠的主要毒性靶器官为肝脏。

Abstract: To explore the acute toxicity of aloperine in mice and to determine the median lethal dose (LD50)and the potential target organs.The mice were intraperitoneally administered with aloperine at the concentrations of 600.00,480.00,384.00,307.20,245.76 mg/kg and observed for toxic manifestations,respectively.LD50 was determined by Bliss method.The survived mice were observed for toxic manifestations for 7 d.The main organs of dead mice (heart,liver,spleen,lung and kidney) were dissected immediately for pathological observation.The LD50 value of aloperine in mice was 395.62 mg/kg (95% confidence interval 305.23 ~ 535.04 mg/kg) with theLD50 value of 143.00 mg/kg and LD95 valueof 1094.47 mg/kg.Liver might be the target organ which was heavily damaged.The work provides experimental basis for further research.
