柴达木枸杞多糖和还原糖含量分析

Analysis of Polysaccharides and Reducing Sugarsin Qaidam Chinese wolfberry

摘要: 建立柴达木枸杞还原糖提取和含量测定的方法,并对不同产地、采摘期柴达木枸杞多糖和还原糖含量进行分析。采用改进的苯酚硫酸法测定多糖的含量,采用优化的3,5二硝基水杨酸法(DNS法)测定还原糖的含量。结果表明:DNS法测定还原糖的最佳条件为:检测波长550 nm,DNS用量3.0 mL,显色时间3 min。多糖含量在2.183~3.863%之间,还原糖含量在49.39~65.15%之间。通过SPSS软件统计分析得出不同产地、采摘期柴达木枸杞的多糖和还原糖含量差异不显著(P>0.05)。DNS法简便快捷,且干扰较小,适于柴达木枸杞还原糖含量测定;通过不同产地、采摘期枸杞多糖和还原糖含量比较分析,为柴达木枸杞品质评价和质量控制提供依据。

Abstract: The objective of this study was to establish a method for extraction and determination of reducing sugars in Qaidam Chinese wolfberry.Meanwhile,the contents of polysaccharides and reducing sugars in wolfberry samples from different habitats and different picking periods were analyzed.The developed phenolsulfuric acid method and optimized 3,5dinitrosalicylicacid (DNS) method were used to analysis the contents of polysaccharides and reducing sugars,respectively.The results show that the optimum parameters of DNS method for reducing sugars are as follows:wavelength was 550 nm,the dosage of DNS solution was 3.0 mL and the color reaction time was 5 min.The contents of polysaccharides and reducing sugars was 2.1833.863% and 49.3965.15%,respectively.The contents of polysaccharides and reducing sugars in Qaidam Chinese wolfberry in different habitats and different picking periods didn’t display significant effect (P> 0.05).The developed method was simple,fastand little interference,and is suitable for the determination of reducing sugars.By comparing analysis of polysaccharides and reducing sugars in Qaidam Chinese wolfberry from different habitats and picking periods,this study effectively provide evidence for the quality evaluation and quality control of Qaidam Chinese wolfberry.
