臭灵丹中三种有效成分提取方法优化及含量测定

Optimization for Extraction of Three Active Ingredients from Laggerae Herba and Content Determination by HPLC

摘要: 在单因素实验的基础上,采用L9(34)正交实验,选择提取时间、料液比、溶剂体积分数3个主要因素,研究各因素交互作用对臭灵丹中冬青酸、洋艾素、臭灵丹酸的提取率的影响,优化臭灵丹中三种有效成分的超声提取方法。臭灵丹中冬青酸、洋艾素、臭灵丹酸的超声法提取最佳工艺参数是,提取时间:40 min;料液比:1:4(g:mL);溶剂体积分数:90%。采用此工艺对云南省德宏芒市产、3~12月份采摘的臭灵丹中三种有效成分进行提取,并以HPLC测定它们的含量,结果表明,9批同一地点、不同时间臭灵丹中三种成分含量差别较大,三种有效成分的含量均在11月份最高。该方法准确度、精密度、稳定性均良好,可用于臭灵丹中有效成分的提取、测定,并指导药材最佳采摘时间。

Abstract: On the basis of single factor experiments,using L9(34) orthogonal experiment,three main factors,extraction time,ratio of solid to liquid,fraction of solution volume are chosen to optimize the ultrasonic extraction method for three main component,Ilicic acid、Artemitin、Pterodontic acid,using the recovery of Ilicic acid,Artemitin,Pterodontic acid as standards.The results show that the optimum extraction process are as follow:the extraction time was 40 min,the ratio of raw material to solvent was:1:4 (g:mL),the fraction of solution volume was 90%.The method was used to treat Laggerae Herba from Dehong Mangshi,Yunnan province,from April to December,and then the content of these three active ingredients was determined by HPLC.The results showed that the content of these three active ingredients varied,at same place but different collecting time.All three active ingredients got the highest content in November.The method is accuracy,precision and stability.It can be used to extract and determine main ingredient from Laggerae Herba,instruct the optimum collecting time as well.
