隔山牛皮消、耳叶牛皮消和戟叶牛皮消中四个C21-甾体苷元含量比较研究

Comparison of Contents of Four C21-Steroidals from Cynanchum wilfordii, Cynanchum auriculatum and Cynanchum bungei

摘要: 传统中药或民间应用对隔山牛皮消(Cynanchum wilfordii)、耳叶牛皮消(Cynanchum auricutalum)和戟叶牛皮消(Cynanchum bungei)三种药材基元植物基本上做白首乌或隔山消混用,其主要活性成分为C21-甾体类成分。三种药材均含有告达亭(caudatin)、去乙酰萝藦苷元(deacymetaplexigenin)、开德苷元(kidjolanin)和加加米宁(gagamine),本文利用HPLC分析技术,检测四个C21-甾体化合物在三种药材中的含量,为合理使用三种药材提供依据。平均每克药材中C21-甾体含量(mg/g)研究结果表明:告达亭在隔山牛皮消中含量(水解前/水解后=0.3542/9.7142)最高、耳叶牛皮消(水解前/水解后=0.2577/4.0825)次之、戟叶牛皮消(水解前/水解后=0.0734/0.1895)最少;其次开德苷元在耳叶牛皮消中含量(水解前/水解后=0.1102/0.6094)比戟叶牛皮消(水解前/水解后=0.0751/0.3225)和隔山牛皮消(水解前/水解后=0.0736/0.2813)高,而后两者含量相近;加加米宁、去乙酰萝藦苷元在三种药材中含量差别不明显。

Abstract: Three plants,Cynanchum wilfordii,Cynanchum auriculatum and Cynanchum bungei,have been used for traditional Chinese medicine as "Bai-Shou-Wu" or "Ge-Shan-Xiao" for a long time,in which the one of main bioactive components are C21-steroids.Since all of three plants contain caudatin,deacymetaplexigenin,kidjolanin,gagamine and their glycosides,which can be used as the standard components for qualitative and quantitative controls of these plants.In this study,HPLC analytic technology was used to detect the contents of the four C21-steroids,providing the method of qualitative and quantitative controls guiding the scientific usage of these medicinal herbs.The average content of C21-steroids in each gram of crude drug (mg/g) was investigated.As the results,the content of caudatin in C.wilfordii (before hydrolysis/after hydrolysis=0.3542/9.7142) was highest than other two species (before hydrolysis/after hydrolysis=0.2577/4.0825 for C.auriculatum,and before hydrolysis/after hydrolysis=0.0734/0.1895 for C.bungei ),while the content of kidjolanin in C.auriculatum (before hydrolysis/after hydrolysis=0.1102/0.6094) was highest one among three species (before hydrolysis/after hydrolysis=0.0751/0.3225 for C.bungei,and before hydrolysis/after hydrolysis=0.0736/0.2813 for C.wilfordii).However,the contents of gagamine and deacymetaplexigenin in the three species were not obvious difference.
