氧化苦参碱对SGC7901与ECV304的体外活性比较研究

摘要: 本论文主要探讨氧化苦参碱对人脐静脉内皮细胞ECV304和人胃癌细胞SGC-7901细胞活性影响并进行比较研究。采用MTT法、生长曲线法检测氧化苦参碱在不同浓度、不同时间对两种细胞增殖抑制作用。HE染色、透射电镜观察氧化苦参碱作用后两种细胞形态变化。结果显示,氧化苦参碱对SGC-7901,ECV304的IC50值分别是660.07、1555.17 μg/mL,对SGC-7901具有明显的增殖抑制作用。氧化苦参碱对SGC-7901细胞形态改变较大,出现细胞质浓缩,细胞染色加深,细胞核变大等形态学改变,细胞出现中期及晚期凋亡。而氧化苦参碱对ECV304形态改变较小,细胞多处于早期凋亡。氧化苦参碱对SGC-7901和ECV304均具有增殖抑制和促凋亡作用,对SGC-7901更明显对,氧化苦参碱具有抗肿瘤细胞增殖、抗肿瘤血管生成作用,但对肿瘤细胞较为敏感。

Abstract: The key point is observing the effects of oxymatrine on the activity of human umbilical vein endothelial cells (ECV304) and human gastric cancer cells(SGC-7901).MTT method and growth curve were used to detect the proliferation inhibitory effect of two kinds of cells at different concentrations and different times,respectively.HE staining and transmission electron microscopy were used to observe the morphological changes of two kinds of cells.The IC50 value of oxymatrine on SGC-7901 and ECV304 were 660.07 μg/mL and 1555.17 μg/mL respectively.SGC-7901 cells ofoxymatrine group appeared significant morphological changes,such as cytoplasm concentration,nucleus getting larger,and a number of cells were in the mid and late apoptotic stage.However,the morphological changes of oxymatrine on ECV304 were not obvious,and some cells were in the early apoptotic stage.Hence,oxymatrine can inhibit the proliferation of the two kinds of cells,but to SGC-7901 cells was more obvious.The morphological changes of SGC-7901 cells were more significant than ECV304 cells at the same concentration.Oxymatrine had obvious anti-tumor activity and partial anti-angiogenesis activity,which indicated that oxymatrine appeared different sensitivity to different cells.
