摘要:

本研究旨在考察白皮杉醇(piceatannol,PIC)对子宫内膜癌细胞HEC-1A的初步抗肿瘤作用。分别通过CCK-8实验、平板克隆形成实验、流式细胞术、划痕损伤修复实验和Transwell小室实验检测不同浓度的PIC对HEC-1A细胞增殖、凋亡、迁移和侵袭能力的影响。Western blot实验检测细胞增殖相关蛋白和侵袭迁移相关蛋白的表达水平。结果显示,与空白组相比,PIC能显著抑制HEC-1A细胞增殖、克隆形成能力、诱导其凋亡并减少细胞迁移距离和侵袭细胞数(P<0.01),下调p-Akt、p-Erk1/2、p-p38MAPK、β-catenin、CD44和Slug蛋白的表达水平,上调E-cadherin蛋白的表达水平(P<0.01)。综上所述,PIC呈浓度依赖性抑制子宫内膜癌HEC-1A细胞的增殖、迁移与侵袭,其机制可能与抑制AKT/ERK/MAPK信号通路和影响Wnt/β-catenin信号通路和上皮-间质转化标志物的改变有关。

Abstract:

The purpose of this study was to investigate the preliminary antitumor effects of piceatannol (PIC) on endometrial cancer cells HEC-1A.The effects of different concentrations of PIC on the proliferation,apoptosis,migration,and invasion abilities of HEC-1A cells was detected by CCK-8 test,plate cloning experiment,flow cytometry,wound healing assay,and Transwell assay.Western blot was used to detect the expression level of cell proliferation related proteins and invasion migration related proteins.The results showed that compared with the control group,PIC significantly reduced the migratory distance and number of invading cells (P<0.01),inhibited the proliferation and clonogenic ability of HEC-1A cells,induced apotosis (P<0.01),and down-regulated the expression levels of p-Akt,p-Erk1/2,p-p38MAPK,β-catenin,CD44 and Slug proteins,and up-regulated the expression levels of E-cadherin protein (P<0.01).In conclusion,PIC demonstrated concentration-dependent suppression of endometrial cancer HEC-1A cell proliferation,migration and invasion.The mechanism may be connected to the inhibition of the AKT/ERK/MAPK signaling pathway and impacting the Wnt/β-catenin signaling pathway and alterations in epithelial-mesenchymal transition markers.
