酸化环境对血管平滑肌细胞(VSMC)增殖的影响及姜黄素对此的作用

Effect of Acidification Environment on Proliferation of Vascular Smooth Muscle Cells and the Influence of Curcumin on It

摘要: 探讨姜黄素对缺血缺氧引起细胞外环境酸化下血管平滑肌增殖的影响及可能机制。在pH 6.9的环境下体外培养大鼠胸主动脉血管平滑肌细胞(VSMC),分别以姜黄素、PCTX-1等处理细胞,并设置空白对照组;采用Western-Blot法测定ASICS 1a蛋白及磷酸化ERK1/2蛋白表达,CCK-8法测定各处理组对VSMC增殖影响。结果显示pH 6.9组显著刺激VSMC的ASICS 1a和ERK1/2磷酸化表达,并促使VSMC增殖;而姜黄素显著抑制大鼠胸主动脉血管平滑肌细胞上ASICS 1a及细胞内磷酸化ERK1/2蛋白表达,从而抑制VSMC增殖。在缺血缺氧所致酸化环境下,大鼠胸主动脉血管平滑肌上的ASIC 1a表达增多,引起细胞内ERK1/2磷酸化,从而对血管平滑肌的增殖进行调控,而姜黄素则表现出抑制作用。

Abstract: To explore the effect and possible mechanism of curcumin on proliferation of vascular smooth muscle in the acidification environmental caused by ischemic hypoxia.The rats' thoracic aorta VSMC cells were cultured in vitro in pH 6.9,treated with curcumin and PCTX-1 respectively,and the blank control group was set.The expression of ASICS 1a protein and phosphorylation ERK1/2 protein was measured by Western-Blot assay,the effect of each treatment group on the proliferation of VSMC was measured by CCK-8 method.The results showed that the pH 6.9 group can significantly stimulate the expression of ASICS 1a and phosphorylation of ERK1/2 in VSMC and promote the proliferation of VSMC.However,curcumin can significantly inhibit the expression of ASICS 1a and intracellular phosphorylation of ERK1/2 protein in the vascular smooth muscle cells of rats' thoracic aorta,thereby inhibiting the proliferation of VSMC.In the acidification environment caused by ischemia hypoxia,the expression of ASICS 1a protein on rats' thoracic aortic vascular smooth muscle increased,caused ERK1/2 phosphorylation in cells,managed the proliferation of vascular smooth muscle,while curcumin showed the inhibitory effect.
