姜黄素药物代谢动力学评价与抗卵巢癌药效作用模拟分析

摘要:

为确定姜黄素药代动力学参数和抗卵巢癌药效机制,我们利用SwissADME、STITCH、DisGeNET、STRING、DAVID v 6.8等数据库分析药代动力学参数,预测抗卵巢癌靶基因,构建靶蛋白互作网络,分析关键靶点KEGG通路。发现姜黄素符合类药性五原则,有一定的胃肠吸收性,但不能通过血脑屏障,且非P-糖蛋白底物。姜黄素通过调控TP53CASP3EGFRERBB2AKT1MTORSTAT3MMP2MMP9HIF1APTGS2CCND1等17个关键卵巢癌靶基因,通过调控肿瘤血管生成、肿瘤浸润转移、肿瘤细胞凋亡、肿瘤免疫、雌激素通路、肿瘤细胞增殖发展等6大类16条信号通路,发挥多途径抗卵巢癌的药理作用。

Abstract:

To determine the pharmacokinetic parameters and anti-ovarian cancer effect of curcumin ,we predicted target genes against ovarian cancer of curcumin by STITCH database,DisGeNET database etc and we constructed and analyzed the protein-interactions network and key target KEGG pathways.We discovered that curcumin was consistent with the Five Principles of drug-like and property of gastrointestinal absorption,but was not consistent with the properties of blood-brain barrier and non-p-glycoprotein substrate.Curcumin play a multi-pathway anti-ovarian cancer pharmacological role via regulates 17 key ovarian cancer target genes included TP53, CASP3,EGFR,Erbb2,KT1,MTOR,STAT3,MMP2,MMP9,HIF1A,PTGS2,CCND1etc. and by regulating tumor angiogenesis,tumor invasion and metastasis,tumor cell apoptosis,tumor immunity,estrogen pathway,tumor cell proliferation and development of 6 categories of 16 signal pathway.
