Innate immune antagonism of rotavirus NSP1 and VP3

摘要: 轮状病毒是引起儿童胃肠炎的重要原因,也是呼肠孤病毒科分节双链RNA病毒家族的一员,编码至少两种宿主天然免疫的直接拮抗剂:NSP1和VP3。NSP1是一种推测的E3泛素连接酶,它介导参与IFN诱导和下游信号转导的细胞因子的降解。VP3是病毒的盖帽酶,它利用一个2H-磷酸二酯酶结构域来阻止细胞内寡腺苷酸合成酶/核糖核酸酶L通路的激活。通过计算、分子和生化研究揭示了NSP1和VP3的天然免疫拮抗作用提供了重要的结构和机制基础。

Abstract: Rotavirus is an important cause of gastroenteritis in children and a member of the segmented double-stranded RNA virus family of reovirus family,encoding at least two direct antagonists of host innate immunity:NSP1 and VP3.NSP1 is a putative E3 ubiquitin ligase,which mediates the degradation of cytokines involved in IFN induction and downstream signal transduction.VP3 is a capping enzyme of the virus.It uses a 2H-phosphodiesterase domain to prevent the activation of intracellular oligoadenylate synthase/ribonuclease L pathway.Computational,molecular and biochemical studies have revealed that the innate immune antagonism of NSP1 and VP3 provides an important structural and mechanism basis.
