川麦冬须根总黄酮和总皂苷提取和富集工艺研究

Optimization on extraction and enrichment technology of total flavonoids and total saponins from fibrous roots of Ophiopogon japonicas

摘要: 本研究采用响应面法优化川麦冬须根中总黄酮和总皂苷的提取工艺。在单因素试验的基础上,以乙醇浓度、料液比、提取时间和提取次数为自变量,以黄酮和皂苷的总得率为响应值,设计四因素三水平Box-Behnken响应面试验优化工艺条件。再以吸附及解吸效果为指标筛选最优的大孔树脂,对总黄酮和总皂苷进一步富集。结果表明,最优提取工艺为:乙醇浓度86%、料液比1∶15、提取时间1.4 h、提取3次,在此条件下浸膏得率26.17%,黄酮和皂苷总得率为2.41%(其中总黄酮得率0.46%,总皂苷得率1.95%)。D101树脂对麦冬须根黄酮和皂苷有较好的富集效果,富集后总黄酮和总皂苷回收率达到88.69%和90.28%,纯度相比浸膏分别提升7倍和9倍。该工艺简单且稳定性好,对于川麦冬须根作为食品资源的开发利用具有重要的参考价值。

Abstract:

This study was to optimize the process of extracting total flavonoids and total saponins simultaneously from fibrous roots of Ophiopogon japonicas using response surface methodology. On the basis of single factor experiment,ethanol volume fraction,solid-liquid ratio,extraction time,and extraction times were selected as independent variables,the overall yield of flavonoids and saponins was used as the response value. Four factors and three levels tests were designed to optimize process conditions by the Box-Behnken experiment. Besides,the optimum macroporous resin was screened in the enrichment stage using adsorption and desorption effect as index. The results showed that the optimal extraction condition was ethanol concentration of 86%,solid-liquid ratio of 1∶15,extraction time of 1.4 h,and extracted three times. The extraction rate was 26.17%,and the total yield of flavonoids and saponins was 2.41% (The yield of total flavonoids and total saponins was 0.46% and 1.95%,respectively). The D101 resin revealed a good ability to enrich flavonoids and saponins. After enrichment,the recovery rate of total flavonoids and total saponins was 88.69% and 90.28%,the content of total flavonoids and total saponins was 7 times and 9 times higher than the crude extract,respectively. The process was simple and stable. It has important reference value for the development and utilization of the fibrous roots of O. japonicas as food resources.
