唐古特白刺多糖对小鼠巨噬细胞RAW 264.7的激活作用及机制研究

Activation of RAW 264.7 macrophages by the polysaccharide from the fruits of Nitraria tangutorum Bobr.and its molecular mechanisms

摘要:

本文采用水提醇沉方法提取得到唐古特白刺(Nitraria tangutorum Bobr.)粗多糖,采用savage法除蛋白,大孔树脂除色素得到白刺精致粗多糖(NTP),利用DEAE-Sepharose FF凝胶柱层析,分离得到3种主要成分NTP1、NTP2及NTP3。并采用PMP柱前衍生化法测定其单糖组成。考察了三种白刺多糖组分对小鼠巨噬细胞RAW 264.7的细胞活力、吞噬功能和NO分泌的影响,并对其炎症及氧化应激通路相关蛋白的表达进行了研究。结果表明,NTP1、NTP2及NTP3的单糖组成以葡萄糖为主,半乳糖或葡萄糖醛酸其次,此外还含有少量其它单糖。NTP1、NTP2及NTP3能够提高RAW 264.7细胞的吞噬能力和NO分泌量。NTP1能够增加氧化应激通路相关蛋白Nrf2及HO-1的表达,而NTP2和NTP3能够激活炎症通路相关的蛋白NF-κB及IκBα。因此可初步判断白刺多糖通过上调Nrf2/HO-1蛋白表达及激活NF-κB通路相关蛋白激活小鼠巨噬细胞RAW 264.7。

Abstract:

The crude polysaccharide was extracted from Nitraria tangutorum Bobr.and precipitated by ethanol then refined by purification through static adsorption and separation.Three main components NTP1,NTP2,NTP3 were achieved by DEAE-Sepharose FF gel filtration chromatography.The composition of monosaccharides determined by PMP pre-column derivation method.In addition,the effects on cell viability,neutral red phagocytoses and NO secretion of RAW 264.7 was carried out.And then,the protein activation/expression of inflammation and oxidative stress signaling pathway were explored.The monosaccharides composition of three NTPs′ monosaccharides consisting mainly of glucose,followed by galactose and glucuronic acid,in addition of few other monosaccharides.NTPs can increase phagocytosis and NO secretion,while NTP1 could activate oxidative stress pathway.NTP2 and NTP3 could activate inflammation pathway.Therefore,the polysaccharides from Nitraria tangutorum Bobr.elicited an activity effect of RAW 264.7 macrophage.
