响应面法优化碱法提取狐臭柴叶多糖工艺及生物活性研究

Optimization of alkali extraction process of polysaccharides from Premna puberula Pamp leaves by response surface methodology and its bioactivity

摘要:

优化狐臭柴叶碱溶性多糖(PLAP)提取工艺,探讨PLAP的体外抗氧化和抗肿瘤活性。采用狐臭柴叶经酸提取法提取的残渣为原料,在单因素基础上,利用响应面优化PLAP的提取工艺,通过抗氧化实验和抗肿瘤实验对PLAP的体外生物活性进行研究。结果表明:PLAP最佳提取工艺为:料液比1∶50 g/mL、NaOH浓度为0.25 mmol/L、提取时间110 min、此条件下PLAP提取率为(11.80±0.50)%。PLAP有较强的抗氧化活性,对DPPH自由基、羟基自由基和ABTS自由基清除率IC50值分别为12.38、3.22和0.18 mg/mL。同时细胞实验表明,PLAP在一定范围内抑制A549细胞和HepG2细胞的增殖与迁移,呈现剂量效应关系。本文为狐臭柴资源的充分开发、临床应用提供理论依据。

Abstract:

The extraction process of alkali-soluble polysaccharide from Premna puberula Pamp leaves(PLAP) was optimized,and the antioxidant and anti-tumor activities activitys of PLAP in vitro was investigated.The residue extracted by acid extraction method was used as raw material.On the basis of single factor,the extraction process of PLAP was optimized by response surface methodology.The in vitro biological activity of PLAP was studied by antioxidant experiments and anti-tumor experiments.The results showed that the optimum extraction process of PLAP was as follows:the ratio of material to liquid was 1∶50 g/mL,the concentration of NaOH was 0.25 mmol/L,and the extraction time was 110 min.Under this condition,the extraction rate of PLAP was (11.80±0.50)%.PLAP had strong antioxidant activity,and the IC50 values of DPPH,hydroxyl and ABTS radicals scavenging rate were 12.38,3.22 and 0.18 mg/mL,respectively.At the same time,cell experiments showed that PLAP could to some range inhibit the proliferation and migration of A549 cells and HepG2 cells,showing a certain dose-effect relationship.This paper provide a theoretical basis for the full development and clinical application of P. puberula resources.
