北美红杉茎发酵液的提高皮肤屏障和抗炎功效研究

摘要:

探讨不同发酵菌种对北美红杉(Sequoia sempervirens)茎发酵液中活性成分的影响,并评估其对提高皮肤屏障及抗炎活性的作用。本研究以北美红杉茎皮为原料,测定不同菌种发酵后的活性成分含量,选择最优菌种组合发酵制得的发酵液进行细胞毒性测试、细胞划痕实验、透明质酸酶活性抑制、炎症因子释放抑制、自由基清除以及抗斑马鱼尾鳍皱缩实验,来验证其提高皮肤屏障及抗炎功效。研究表明,酿酒酵母菌(Saccharomyces cerevisiae)1-1与枯草芽孢杆菌(Bacillus subtilis)2-1的复合发酵系统能够显著提高发酵液中活性成分的含量。该发酵液在体外实验中表现出显著的透明质酸酶抑制作用,其IC50值为9.89 mg/mL,并对DPPH自由基、超氧阴离子自由基和羟基自由基的清除能力较强。此外,细胞实验结果显示,中低浓度的发酵液对HaCaT细胞无明显毒性作用,在10 mg/mL时,能显著促进HaCaT细胞的伤口愈合。进一步研究发现,10 mg/mL浓度的北美红杉茎发酵液能够有效抑制脂多糖刺激下巨噬细胞分泌肿瘤坏死因子-α和白细胞介素-6的水平。最后,通过抗斑马鱼尾鳍皱缩实验验证了该发酵液对抗紫外线诱导的光老化具有显著作用。本研究提出了一种高效制备北美红杉茎皮活性物质的方法,并证明其发酵液在提升皮肤屏障功能和抗炎活性方面具有潜在应用价值,为北美红杉在皮肤护理、修复及抗衰老领域的应用提供了新的研究方向。

Abstract:

This study aims to investigate the effects of different fermentation strains on the active components of the fermentation broth of Sequoia sempervirens stem, and to evaluate its potential in enhancing skin barrier function and anti-inflammatory activity. The bark of Sequoia sempervirens stem was used as a starting material to determine the content of active ingredients after fermentation with different strains. The optimal strain combination was selected to ferment the resulting liquid, which was then subjected to cell toxicity tests, cell scratch assays, hyaluronidase activity inhibition, inhibition of inflammatory factor release, free radical scavenging, and anti-zebrafish tail fin shrinkage experiments to validate its efficacy in enhancing skin barrier function and anti-inflammatory effect.The results showed that the compound fermentation system of Saccharomyces cerevisiae 1-1 and Bacillus subtilis 2-1 significantly increased the content of active ingredients in the fermentation broth. This fermentation broth exhibited remarkable hyaluronidase inhibitory activity in vitro, with an IC50 value of 9.89 mg/mL and showed strong scavenging ability against DPPH free radicals, superoxide anions free radicals, and hydroxyl free radicals. Additionally, the cellular experimental results indicated that low to moderate concentrations of the fermentation broth had no obvious cytotoxic effects on HaCaT cells, while 10 mg/mL significantly promoted the wound healing of HaCaT cells. Further study revealed that the fermentation broth with concentration of 10 mg/mL could effectively inhibit the levels of Tumor necrosis factor-α and Interleukin-6 secreted by macrophages stimulated with lipopolysaccharide Finally, the anti-photoaging effect of the fermentation broth under UV radiation was verified by the zebrafish tail fin wrinkle experiment. This study proposes an efficient method for preparing active ingredients from the bark of Sequoia sempervirens stem and demonstrates the potential value of the fermentation broth in enhancing skin barrier function and anti-inflammatory activity, providing a new research direction for the application of Sequoia sempervirens in skincare, repair, and anti-aging.
