超声辅助深共熔溶剂提取构树叶总黄酮工艺优化及抗氧化活性分析

Optimization of ultrasonic-assisted extraction of total flavonoids from Broussonetia papyrifera leaves by deep eutectic solvent and its antioxidant activity

摘要:

优化构树叶总黄酮(total flavonoids from Broussonetia papyrifera leaves,TFBPL)的超声辅助深共熔溶剂(deep eutectic solvent,DES)提取工艺,并评价其抗氧化活性。在单因素试验的基础上,采用响应面法优化提取工艺,评估其对DPPH、ABTS和羟基自由基的清除能力,探究其体外抗氧化活性。结果表明,甜菜碱-乙醇为DES,摩尔比为1∶3,含水量29.0%(V/V),料液比1∶43(g/mL),超声时间32 min时,TFBPL得率为60.66±0.46 mg/g。在相同条件下,此法的TFBPL得率是传统溶剂提取的1.8~3.2倍。TFBPL在0.5~60 mg/g的质量浓度范围内,对DPPH、ABTS和羟基自由基的清除率分别为72.97%±0.98%、98.53%±0.56%和51.60%±0.82%。因此,超声辅助DES提取TFBPL的工艺高效、环保,其提取获得的TFBPL具有显著的体外抗氧化活性。

Abstract:

This study aims to optimize the ultrasonic-assisted deep eutectic solvent(DES ) extraction process of total flavonoids from Broussonetia papyrifera leaves (TFBPL) and evaluate its antioxidant activity.Based on the single factor experiments,the extraction process was optimized by response surface methodology.The antioxidant activity was evaluated by determining the scavenging capacity of TFBPL against DPPH,ABTS and hydroxyl radicals in vitro.The results revealed that when betaine-ethanol was employed as the DES at a molar ratio of 1∶3,water content of 29.0% (V/V),solid-liquid ratio of 1∶43 (g/mL),and ultrasonic time of 32 min,the yield of TFBPL reached 60.66±0.46 mg/g.Under the same conditions,the yield of TFBPL obtained by this method was 1.8-3.2 times higher than that obtained by traditional solvent extraction.Within a mass concentration range of 0.5 to 60 mg/g,the scavenging rates of TFBPL against DPPH,ABTS and hydroxyl radicals were 72.97%±0.98%,98.53%±0.56% and 51.60%±0.82%,respectively.Therefore,the ultrasonic-assisted DES extraction process for TFBPL is both efficient and environmentally friendly,and the extracted total flavonoids exhibit significant antioxidant activity in vitro.
