喜碱鸢尾根总多酚的提取工艺优化及其对具核梭杆菌抑制活性研究

Optimization of the extraction process of total polyphenols from Iris halophila Pall roots and its inhibitory activity against Fusobacterium nucleatum

摘要:

优化喜碱鸢尾根总多酚提取工艺,并考察喜碱鸢尾根总多酚对具核梭杆菌(Fusobacterium nucleatum,Fn)的体外抑菌活性。首先以喜碱鸢尾根中总多酚含量为评价指标,考察液料比、提取时间和提取次数对总多酚含量的影响;根据单因素试验结果,采用Box-Behnken响应面法优化喜碱鸢尾根总多酚的提取工艺;通过琼脂打孔法和微量稀释法测定不同质量浓度总多酚对Fn的抑制作用和生长过程的影响,结晶紫染色法测定对Fn生物膜形成的影响。结果表明,喜碱鸢尾根总多酚最佳提取工艺为:液料比20∶1(mL/g),提取次数为3次,提取时间85 min,在此条件下喜碱鸢尾根总多酚含量为36.52 mg/g;Fn最佳生长周期为40 h;喜碱鸢尾根总多酚对Fn的最小抑菌浓度为50 mg/mL,最小杀菌浓度为200 mg/mL;当药物浓度为12.5、25、50和100 mg/mL时,Fn生物膜活性分别为59.64%、47.80%、18.58%和4.78%,Fn生物膜活性与喜碱鸢尾根总多酚浓度呈负相关(P<0.001)。本研究优选的提取工艺稳定可行,获得的提取物总多酚含量高,能有效抑制Fn及其生物膜的活性,为合理开发利用喜碱鸢尾根奠定坚实基础。

Abstract:

This study aimed to optimize the extraction process of total polyphenols from Iris halophila Pall roots and evaluate their in vitro inhibitory effects on Fusobacterium nucleatum (Fn).The total polyphenols content in the roots of I.halophila was used as the evaluation index to investigate the effects of liquid-material ratio,extraction time and extraction times;Based on the results of single factor experiments,Box-Behnken response surface methodology was used to optimize the extraction process of total polyphenols from I.halophila roots.The inhibitory effects and growth process of total polyphenols with different mass concentrations on Fn were determined by agar perforation method and microdilution method.Meanwhile the effect on Fn biofilm formation was determined by crystal violet staining method.The results showed that the optimum extraction process of total polyphenols from I.halophila roots was as follows:liquid-material ratio was 20∶1 (mL/g),extraction times was 3,extraction time was 85 min.The yield of total polyphenols from I.halophila roots under these conditions was 36.52 mg/g.The optimal growth period of Fn was 40 h;The minimum inhibitory concentration of total polyphenols from I.halophila roots against Fn was 50 mg/mL,and the minimum bactericidal concentration was 200 mg/mL;When the drug concentrations were 12.5,25,50 and 100 mg/mL,the biofilm activity of Fn was 59.64%,47.80%,18.58%,and 4.78% respectively.The biofilm activity of Fn was negatively correlated with the concentration of total polyphenols from I.halophila roots (P<0.001).The optimized extraction process is stable and feasible,and the high content of total polyphenols show effective activity in inhibiting Fn and its biofilm,which lays a solid foundation for the rational development and utilization of I.halophila roots.
