蚂蚁黑色素的提取、纯化及抗氧化活性研究

Extraction, Purification and Antioxidant Activity of Melanin from the Black Ants

摘要: 对黑蚂蚁采用盐酸水解、氢氧化钠溶液提取、盐酸沉淀,以及多种有机溶剂洗涤等工艺手段提取分离、纯化得到黑蚂蚁黑色素。研究发现黑蚂蚁黑色素溶于碱性溶液,不溶于水、酸及有机溶剂;其紫外吸收光谱与红外光谱具有吲哚型黑色素典型光谱性质。利用分光光度法测定了黑蚂蚁黑色素的总抗氧化能力、羟基自由基清除能力及还原力,试验结果表明:黑蚂蚁黑色素有一定的抗氧化能力,但在试验范围内其抗氧化能力比维生素C低。

Abstract: The melanin derived from the black ants was isolated and purified by acid hydrolysis, alkaline extraction, acid deposition and organic solvents abstersion. The solubility showed that the melanin was soluble in alkaline solution, but insoluble in water, acid solution and organic solvent. It may be a type of indoles melanin from the UV-visible spectrogram and the infrared spectrogram. The total antioxidant capacity, hydroxyl radical scavenging capacity and reducing power of the melanin were measured by the spectrophotometric method. The results showed that the melanin had certain antioxidant activity, but lower than that of the vitamin C.
