苦参内生真菌BS002菌株有效代谢产物的分离及结构鉴定

摘要: 本论文采用硅胶柱层析的方法,以苦参内生真菌BS002菌株为研究对象,对BS002菌株的有效代谢产物进行了分离,并最终得到两种有效活性成分。论文确定了有效产物提取分离的最适温度为40 ℃、pH为5。柱层析分离选用硅胶(GF254)为固定相,CHCl3:CH3OH=5:1为流动相进行,并经TLC生物自显影法跟踪生物活性物质,最终产物经浓缩、冻干,利用HPLC、IR、GC-MS等方法确定其分别为:1,2-苯二甲酸丁基环己基酯,1,2-苯二甲酸-单(2-乙基己基)酯。

Abstract: Silica gel column chromatography was used to isolate active metabolites from fermentation liquid of endophytic fungus BS002 isolated from S.flavescens, and eventually two effective active ingredients were obtained in this paper.The optimal temperature 40 ℃ and pH 5 were determined to separate effective product.GF254 was selected as the stationary phase,and CHCl3:CH3OH = 5:1 was selected as the mobile phase in the test while bioactive metabolites were tracked by TLC. Biological active ingredients were separated,concentrated and lyophilized,then identified to be 1,2-Benzenedicarboxylic acid,mono(2-ethylhexyl) ester and 1,2-Benzenedicarboxylic acid,butyl cyclohexyl ester by methods of HPLC, IR, GC-MS.
