菊苣抗氧化及抗炎活性研究

Antioxidant and anti-inflammatory activities of Cichorium intybus L.

摘要:

为明确菊苣提取物中总多酚的含量及其抗氧化、抗炎活性,本研究以菊苣地上部分、叶以及菊苣加工茶制品为提取原料,用水或乙醇为溶剂进行回流提取,继而采用D101大孔树脂法或溶剂法处理,获得菊苣固体提取物。采用福林酚法测定菊苣提取物中总多酚的含量,采用DPPH法和ABTS法评估各提取物的体外抗氧化活性,采用Griess法和ELISA法评估菊苣提取物对于脂多糖诱导的巨噬细胞RAW 264.7释放NO和白细胞介素-6(interleukin-6,IL-6)的影响。实验结果显示各菊苣提取物中,经过D101树脂柱纯化的菊苣叶提取物(CIE-11)的总多酚的含量最高(506%),抗氧化活性最强(DPPH法,IC50=6.0 μg/mL;ABTS法,IC50=1.835 μg/mL),对于总多酚含量及其抗氧化活性的相关性分析表明二者呈现显著相关。进一步的细胞实验结果显示CIE-11具有显著的抑制LPS诱导的小鼠巨噬细胞RAW 264.7释放NO和IL-6的作用,表明其具有潜在的抗炎药理活性。本研究可为菊苣在抗炎、抗氧化方面的应用及药物研发提供参考依据。

Abstract:

To clarify the total polyphenol content (TPC) and antioxidant/anti-inflammatory activities of Cichorium intybus L.,this study used the aboveground parts,leaves,and processed tea products of C.intybus as raw materials,with water or ethanol as solvents for reflux extraction.The extracts were then processed by D101 macroporous resin method or solvent method to obtain solid extracts.The Folin-Ciocalteu method was employed to determine total polyphenol content,the DPPH and ABTS assays were used to evaluate in vitro antioxidant activities,and Griess and ELISA methods were applied to assess the effects of the extracts on lipopolysaccharide (LPS)-induced nitric oxide (NO) and interleukin-6 (IL-6) release in macrophage RAW 264.7 cells.Experimental results showed that among all the extracts,the D101 resin-purified leaf extract (CIE-11) exhibited the highest total polyphenol content (50.6%) and the strongest antioxidant activity (IC50=6.0 μg/mL by DPPH assay;IC50=1.835 μg/mL by ABTS assay).Correlation analysis between TPC and antioxidant activity revealed a significant positive correlation.Further cellular experiments demonstrated that CIE-11 significantly inhibited LPS-induced NO and IL-6 release in mouse macrophage RAW 264.7 cells,indicating its potential anti-inflammatory pharmacological activity.This study provides a reference for the application of chicory in anti-inflammatory and antioxidant fields,as well as for drug development.
