白术多糖对CUMS模型小鼠抑郁样行为及其脑肠轴和HPA轴的影响

Effects of Atractylodes macrocephala polysaccharides on depressive-like behavior, brain-gut axis and HPA axis in CUMS model mice

摘要:

本研究旨在观察白术多糖(Atractylodes macrocephala polysaccharides,AMP)对动物慢性不可预知应激模型(chronic unpredictable mild stress,CUMS)小鼠抑郁样行为及其脑肠轴和下丘脑-垂体-肾上腺轴(hypothalamic-pituitary-adrenal axis,HPA)的影响,以揭示AMP在抗抑郁方面的多靶点作用。采用CUMS方式建立小鼠抑郁模型,将小鼠随机分为空白组、模型组(CUMS)、AMP低剂量组(2.5 mg/kg)、AMP中剂量组(5 mg/kg)、AMP高剂量组(10 mg/kg)及氟西汀组(0.6 mg/kg)。观察AMP对各组小鼠的行为学指标包括糖水偏好、悬尾静止时长和强迫游泳静止时长;小鼠下丘脑中神经递质包括5-羟色胺(5-hydroxy tryptamine,5-HT)、去甲肾上腺素(norepinephrine,NE)、多巴胺(dopamine,DA)含量;小鼠血清中炎症因子和激素水平包括白细胞介素-6(interleukin-6,IL-6)、促肾上腺皮质激素(adrenocor ticotropic hormore,ACTH)、皮质酮(corticosterone,CORT)含量;小鼠海马区尼氏体神经元细胞;小鼠结肠中嗜铬粒蛋白A(chromogranin A,CgA)表达量等指标的影响。结果表明:AMP组(2.5、5、10 mg/kg)显著改善CUMS抑郁小鼠的各项行为指标(P<0.05);恢复CUMS抑郁小鼠受损的尼氏体(P<0.05);回调小鼠结肠中CgA表达量和5-HT水平(P<0.05)并呈现量效关系。同时,AMP还显著回调CUMS抑郁小鼠脑内5-HT、NE、DA水平(P<0.05),及血清中IL-6、CORT和ACTH的水平(P<0.05)。AMP通过调节脑-肠轴和HPA轴的多靶点作用,展现出对抗抑郁样行为的潜在治疗作用。

Abstract:

This study aims to investigate the effects of Atractylodes macrocephala polysaccharides (AMP) on depression-like behavior,the brain-gut axis,and the hypothalamic-pituitary-adrenal axis (HPA) in chronic unpredictable mild stress (CUMS) model mice,as well as to elucidate the multi-target effects of AMP in combating depression.Mice were randomly assigned to a control group,a model group,a low dose AMP group (2.5 mg/kg),a middle dose AMP group (5 mg/kg),a high dose AMP group (10 mg/kg) and a fluoxetine group (0.6 mg/kg).Behavioral indices influenced by AMP were assessed across all groups,including sucrose preference test,tail suspension test and forced swimming test.Neurotransmitter levels in the hypothalamus included serotonin (5-HT),norepinephrine (NE) and dopamine (DA) were detected.Serum concentrations of inflammatory factors and hormones such as interleukin-6 (IL-6),adrenocorticotropic hormone (ACTH) and corticosterone (CORT) were also evaluated.The expression of chromogranin A (CgA) was analyzed in mouse colon tissues alongside nissl-stained neurons within the hippocampus.Results indicated that AMP at doses of 2.5,5 and 10 mg/kg significantly improved behavioral indices in CUMS-induced depressed mice (P<0.05), and recovery of damaged nissl staining was observed in these subjects as well (P<0.05).Furthermore,both CgA expression levels and serotonin concentrations in mouse colons returned to baseline values following treatment with AMP (P<0.05),demonstrating a dose-dependent relationship.Concurrently,levels of 5-HT,NE,DA within brain tissue along with IL-6,CORT,ACTH serum concentrations among CUMS-depressed mice were markedly reduced by AMP (P<0.05).These findings suggest that AMP has potential therapeutic implications for intervening in depression-like behaviors through modulation of multiple targets within both the brain-gut axis and HPA axis.
