摘要:

刺梨是宝贵的药食两用植物资源,通过研究其不同入药部位黄酮类化合物分布的差异,为刺梨的进一步开发和利用提供理论依据。本研究以贵州刺梨为研究对象,基于非靶向代谢组学技术结合主成分分析、正交偏最小二乘判别分析等方法,对其根、叶、果实中的代谢物进行差异性分析。采用超高效液相色谱-串联质谱(UPLC-MS)技术,结合代谢组学分析方法,对刺梨根、叶、果实中黄酮类成分进行定性与定量分析。结果共检测到3 601种代谢物,其中根、叶、果实分别鉴定出165、192、180种黄酮类化合物,涵盖查耳酮、黄酮、黄酮醇、花青素等9类。主成分分析结果显示,3组样本显著分离,表明不同部位代谢物差异显著。差异代谢物分析(差异倍数≥2或≤0.5,变量重要性投影值≥1)表明,叶与果实、叶与根、果实与根分别存在110、118和82个差异黄酮类代谢物。KEGG富集分析显示差异代谢物主要参与类黄酮生物合成途径(代谢物数占比最高,达6~7个/组),表明其分布与器官特异性功能密切相关。综上,本研究系统揭示了刺梨不同入药部位的黄酮类成分特征,为药用资源开发提供了理论依据。

Abstract:

Rosa roxburghii Tratt.(RRT)is a valuable medicinal and edible plant resource.Investigating the distribution differences of flavonoid compounds in its various medicinal parts provides a theoretical basis for the further development and utilization of RRT.This study focused on Guizhou RRT and employed non-targeted metabolomics combined with principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) to analyze the metabolic differences among its root,leaf,and fruit.In this study,the UPLC-MS technology was adopted,combined with metabolomics analysis methods,to conduct qualitative and quantitative analysis of flavonoids in the root,leaf and fruit of RRT.A total of 3 601 metabolites were detected,among which 165,192 and 180 flavonoids were identified in the root,leaf and fruit respectively,covering 9 types such as chalcones,flavones,flavonols and anthocyanins.The PCA results showed that the three groups of samples were significantly separated,indicating significant differences in metabolites among different parts.The analysis of differential metabolites (fold change≥ 2 or ≤ 0.5,variable importance in projection value≥ 1) indicated that there were 110,118 and 82 differential flavonoid metabolites between leaf and fruit,leaf and root,and fruit and root respectively.KEGG enrichment analysis showed that the differential metabolites were mainly involved in the flavonoid biosynthesis pathway (with the highest proportion of metabolites,reaching 6-7 per group),indicating that their distribution was closely related to the organ-specific functions.In conclusion,this study systematically revealed the characteristics of flavonoids in different medicinal parts of RRT,providing a theoretical basis for the development of medicinal resources.
