基于肠道菌群探讨罗布麻多糖改善小鼠代谢性脂肪性肝病的作用

Effects of Apocynum venetum polysaccharide on improving metabolic associated fatty liver disease in mice based on gut microbiota

摘要:

探讨罗布麻多糖(Apocynum venetum polysaccharide,AVP)对高脂饮食诱导的代谢性脂肪性肝病(metabolic associated fatty liver disease,MAFLD)的治疗效果及其机制。实验将40只6~8周龄雄性C57BL/6小鼠随机分为正常组、模型组、AVP高剂量组(400 mg/kg)和AVP低剂量组(200 mg/kg),采用高脂饮食建立MAFLD模型,干预12周后测定小鼠血清生化指标,测定肝脏组织损伤、氧化应激、脂质含量、炎症因子指标,进行HE染色、油红O染色观察肝脏病理变化,采用16S rRNA基因测序检测小鼠粪便肠道菌群。结果显示,AVP显著改善了小鼠的肝功能指标,降低了血清中的转氨酶水平,并提高了白蛋白水平(P<0.01或P<0.05)。AVP干预还显著改善了小鼠的血脂异常,肝脏组织病理学检查显示,AVP能够减轻脂肪变性、炎症细胞浸润和肝细胞肿大,减轻肝脏炎症反应(P<0.01或P<0.05)。16S rRNA基因测序结果显示,AVP的干预显著增加了有益菌如双歧杆菌(Bifidobacterium)和乳酸杆菌(Lactobacillus)的丰度,同时降低了促炎菌如肠球菌(Enterococcus)的丰度(P<0.01或P<0.05),AVP能够恢复肠道微生物的多样性和丰度。综上所述,AVP通过调节肠道菌群结构,改善了MAFLD的进展,为利用天然植物多糖治疗MAFLD提供了新的思路。

Abstract:

This study aimed to investigate the therapeutic effects and mechanisms of Apocynum venetum polysaccharide (AVP) on high-fat diet-induced metabolic associated fatty liver disease (MAFLD). Forty male C57BL/6 mice aged 6-8 weeks were randomly divided into four groups: normal group, model group, high-dose AVP group (400 mg/kg), and low-dose AVP group (200 mg/kg). A MAFLD model was established using a high-fat diet, followed by 12 weeks of intervention. Serum biochemical indicators, liver tissue injury, oxidative stress, lipid content, and inflammatory cytokine levels were measured. Hepatic pathological changes were observed through HE staining and Oil Red O staining, while 16S rRNA gene sequencing was employed to analyze gut microbiota in fecal samples. The results demonstrated that AVP significantly improved liver function parameters by reducing serum transaminase levels and elevating albumin levels (P<0.01 or P<0.05). AVP intervention also effectively ameliorated dyslipidemia. Histopathological examination revealed that AVP alleviated hepatic steatosis, inflammatory cell infiltration, and hepatocyte swelling, thereby mitigating liver inflammation (P<0.01 or P<0.05). 16S rRNA sequencing analysis indicated that AVP intervention notably increased the abundance of beneficial bacteria such as Bifidobacterium and Lactobacillus, while reducing pro-inflammatory bacteria like Enterococcus (P<0.01 or P<0.05). Furthermore, AVP restored the diversity and abundance of gut microbiota. In conclusion, AVP ameliorates MAFLD progression by modulating gut microbiota structure, providing novel insights for utilizing natural plant polysaccharides in MAFLD treatment.
