摘要:

建立白扁豆和扁豆衣的指纹图谱及竹节参皂苷IVa(chikusetsusaponin IVa,CS-IVa)的含量测定方法,并与黑扁豆、红扁豆及其他17种豆类进行比较,以探究其药用价值。采用高效液相色谱法(HPLC)建立特征图谱,并通过化学计量法进行聚类分析(hierarchical cluster analysis,HCA)、主成分分析(principal component analysis,PCA)和正交偏最小二乘判别分析(orthogonal partial least squares discriminant analysis,OPLS-DA)对不同种类和部位的扁豆及其他豆类进行化学模式识别分析。结果表明,白扁豆和扁豆衣的HPLC特征图谱确定了14个共有峰,其中2个为共有色谱峰(原儿茶酸、大豆苷),2个为差异特征峰(染料木苷、CS-IVa)。HCA将15批样品分为2类,OPLS-DA分析筛选出7个影响质量的共有特征峰,CS-IVa被确定为定量分析指标,且主要集中在种皮部。研究结果表明,白扁豆和扁豆衣在HPLC特征图谱和CS-IVa含量上与其他常见豆类有明显区别,为科学阐述其药用价值提供了依据。

Abstract:

This study aimed to establish the HPLC fingerprint profiles of Lablab Semen Album (LSA) and its seed coat (LSAC),and the content determination method for chikusetsusaponin IVa (CS-IVa),and compare them with Dolichos lablab seed with black seed coat (black lentils) and with red seed coat (red lentils),as well as 17 other types of beans to explore their medicinal value.HPLC was used to establish the characteristic profiles,and chemometric methods including hierarchical cluster analysis (HCA),principal component analysis (PCA),orthogonal partial least squares discriminant analysis (OPLS-DA) were applied to perform chemical pattern recognition analysis on different types and parts of lentils and other beans.The results showed that the HPLC fingerprint profiles of LSA and LSAC were established with 14 common peaks identified,including two common chromatic peaks (protocatechuic acid and soyasaponin) and two differential characteristic peaks (genistin and CS-IVa).HCA classified 15 batches of samples into two categories,and OPLS-DA analysis screened out seven common characteristic peaks affecting quality.CS-IVa was determined as the quantitative analysis indicator and was mainly concentrated in the seed coat.The study results indicated that LSA and LSAC were clearly distinguished from other common beans in terms of HPLC fingerprint profiles and CS-IVa content,providing a scientific basis for elucidating their medicinal value.
