甘松不同部位主要成分分析

摘要:

为探讨甘松不同部位主要成分的差异,挖掘其潜在价值。通过水蒸气蒸馏法测定甘松不同部位的挥发油含量,采用高效液相色谱法和紫外-可见分光光度法测定不同部位的甘松新酮、绿原酸和总黄酮的含量,采用气相色谱-质谱联用技术分析不同部位的挥发性成分,结合正交偏最小二乘判别分析(orthogonal partial least-squares discrimination analysis,OPLS-DA)确定不同部位的关键挥发性差异成分。结果表明甘松根部挥发油(6.00%)和甘松新酮(49.0 mg/g)含量最高,茎部总黄酮(46.7 mg/g)和绿原酸(17.4 mg/g)含量最高。不同部位共鉴定53个挥发性成分,其中根47个,茎35个,叶46个,花50个,倍半萜类是各部位主要挥发性成分。根中倍半萜类成分相对含量最高,茎中醛类和酸类最高,叶中酚类、单萜类、酮类、其他类最高,花中酯类、醇类最高。OPLS-DA结果说明不同部位挥发性成分存在区别,变量重要性投影结果筛选出依兰醛、白菖烯、十六酸等13个关键挥发性差异成分。综上表明,甘松不同部位的主要成分存在差异,为后续甘松不同部位的深入挖掘利用提供科学依据。

Abstract:

This study aims to investigate the differences of main components in different parts of Nardostachys jatamansi DC.and dig its potential value.The content of volatile oil in different parts of N.jatamansi were determined by steam distillation,the content of nardosinone,chlorogenic acid,and total flavonoids in different parts were determined by high performance liquid chromatography and ultraviolet-visible spectroscopy,the volatile components in different parts were analyzed by gas chromatography-mass spectrometry,and the key volatile differential components were identified by orthogonal partial least-squares discrimination analysis (OPLS-DA).The results showed that the content of volatile oil (6.00%) and nardosinone (49.0 mg/g) were the highest in roots,and the content of total flavonoids (46.7 mg/g) and chlorogenic acid (17.4 mg/g) were the highest in stems.A total of 53 volatile components were identified from different parts,including 47 from roots,35 from stems,46 from leaves,and 50 from flowers.Sesquiterpenes were the main volatile components from different parts.The relative content of sesquiterpenes was the highest in roots,the relative content of aldehydes and acids were the highest in stems,the relative content of phenols,monoterpenes,ketones and others were the highest in leaves,and the relative content of esters and alcohols were the highest in flowers.The OPLS-DA results showed that there were differences in volatile components in different parts,and 13 key volatile components such as ylangenal,calarene,and hexadecanoic acid were screened out by variable importance in projection results.In conclusion,there are differences in the main components of different parts from N.jatamansi,which can provide a scientific basis for the further excavation and utilization of different parts of N.jatamansi.
