秋鼠曲草总黄酮提取及纯化工艺研究

Extraction and Purification Technology of Total Flavonoids from Gnaphalium hypoleucum D.C.

摘要: 以乙醇浓度、固液比、提取时间为考察因素,以总黄酮提取量为指标,采用单因素试验和正交试验优选秋鼠曲草总黄酮提取工艺。以总黄酮的吸附率、解吸率为指标,优选纯化秋鼠曲草总黄酮的大孔吸附树脂,并对其动态吸附与解吸工艺进行筛选,确定纯化秋鼠曲草总黄酮的工艺参数。确定最佳提取工艺为70%乙醇,固液比1:20,提取2次,每次1h,秋鼠曲草中总黄酮含量为10.47%。X5型大孔树脂对秋鼠曲草总黄酮具有最佳的吸附分离性能,纯化工艺条件为:秋鼠曲草总黄酮上样质量浓度为2.18~2.91 mg/mL,上样体积流量为1 mL/min,最大上样量以总黄酮计为26.19 mg/g树脂;洗脱剂为30%乙醇,洗脱流速为3 mL/min,洗脱剂用量为25BV,其30%乙醇洗脱物中总黄酮质量分数提高到42.41%,总黄酮回收率达84.05%。

Abstract: The extract technology of the total flavonoids from Gnaphalium hypoleucum D.C.was optimized by orthogonal experiment with the yield of total flavonoids from G.hypoleucum as index,and the ethanol concentration,extracting time,the liquidsolid ratio as factors.The optimal extraction technology was as follows:70% ethanol,reflux extraction 2 times,each for 1 hour with liquidsolid ratio of 1:20.The content of total flavonoids was 10.74%.The enrichment and purification technology of total flavonoids with macroporous resin.Static adsorption and dynamic adsorption were used to optimize the adsorption and elution process by macroporous resin.X5 resin could effectively enrich the total flavonoids,the optimal conditions were:liquid concentration of 2.182.91 mg/mL,absorption velocity 1 mL/min,resin adsorption quantity 26.19 mg/g,eluting with 30% ethanol for 25BV,elution speed 3 mL/min.Under this condition,the content of total flavonoids increased to 42.41%,and the desorption rate was 84.05%.
