体外降胆固醇活性柠檬内生菌的分离筛选及鉴定

Isolation,screening and identification of lemon endophyte with high cholesterol-reducing activity in vitro

摘要:

为了提高胆固醇降解率,筛选高降脂活性发酵菌剂的优良菌种。采用连续稀释平板法结合高效液相色谱法,分离柠檬内生菌并对其降胆固醇活性进行筛选,再采用16S或26S rDNA序列分析进行活性菌种鉴定。研究结果表明从柠檬中分离得到122株柠檬内生菌,初筛得到23株胆固醇降解率高于25%的菌株,进一步进行复筛得到17株降胆固醇活性菌株,经鉴定结果为酵母属16株,其中Meyerozyma carpophila 7株,Meyerozyma guilliermondii 5株,Wickerhamomyces anomalusPichia membranifaciens各2株,细菌1株鉴定为Acetobacter ghanensis。该研究菌株可作为发酵剂,为降脂类益生菌功能产品的开发提供基础。

Abstract:

To improve the efficiency of cholesterol-lowering ability in vitro and screen the excellent strains for fermentation.The serial dilution plate method was used to isolate cholesterol-reducing strains from Citrus limon.The activity of cholesterol-reducing was determined by HPLC,and then identified by 16S or 26S rDNA sequence analysis.A total of 122 strains were isolated from Citrus limon,and 23 cholesterol-lowering strains were preliminarily screened out firstly,then 17 strains showed activity by further determination.Among them,16 strains belonged to Saccharomycetales sp.,seven strains were Meyerozyma carpophila,five strains were Meyerozyma guilliermondii,four strains were Wickerhamomyces anomalus and Pichia membranifaciens.One strain was Acetobacter ghanensis. These strains can be used as the starter culture,which has provided a basis for the development of probiotics functional products.
