辣椒素影响CUMS抑郁大鼠摄食行为的机制研究

Mechanism of capsaicin in modulating feeding behavior of CUMS-induced depressed rats

摘要:

探究辣椒素(capsaicin,CAP)改善慢性不可预测轻度应激(chronic unpredictable mild stress,CUMS)抑郁大鼠的摄食行为机制。利用随机数字表法将40只大鼠随机分为正常对照组和模型组,其中正常对照组10只,模型组30只,通过CUMS方法制备抑郁模型,将造模成功的30只大鼠随机分为模型组、辣椒素组(0.05 mg/kg)和氟西汀组(10 mg/kg),每组各10只,所有大鼠均接受腹腔注射,持续4周。采用Western blot法检测下丘脑弓状核(arcuate nucleus,ARC)组织中神经肽Y(neuropeptide Y,NPY)、刺鼠关联蛋白(agouti-related protein,AgRP)、大麻素受体1型(cannabinoids receptor 1,CB1R);免疫荧光检测NPY/CB1R和AgRP/CB1R共表达;采用实时荧光定量PCR检测ARC组织中γ-氨基丁酸A受体(γ-aminobutyric acid A receptor,GABAAR)和γ-氨基丁酸B受体(γ-aminobutyric acid B receptor,GABABR) mRNA表达。结果显示,与正常对照组比较,模型组NPY/AgRP/CB1R蛋白表达均降低(P<0.01,P<0.001),NPY、AgRP分别与CB1R荧光共定位降低(P<0.05;P<0.001),模型组GABAAR mRNA表达降低,GABABR mRNA表达升高(P<0.05,P<0.01);与模型组比较,给药组NPY/AgRP/CB1R蛋白表达均升高(P<0.01,P<0.001),NPY、AgRP分别与CB1R荧光共定位强度升高(P<0.01,P<0.001),GABAAR mRNA表达升高(P<0.05),GABABR mRNA表达降低(P<0.05,P<0.01)。以上结果表明,辣椒素能够改善CUMS抑郁大鼠的抑郁行为及摄食行为,其机制可能通过上调CUMS抑郁大鼠ARC脑区NPY/AgRP/CB1R/GABAAR表达及下调GABABR受体表达发挥调控作用。

Abstract:

This study aims to investigate the mechanism of capsaicin (CAP) to improve the feeding behavior of chronic unpredictable mild stress (CUMS) depressed rats. Forty rats were randomly divided into normal group and model group using random number table method, among which ten rats were in normal group and 30 rats were in model group. The depression model was prepared by CUMS method, and the 30 rats with successful modeling were randomly divided into the model group, capsaicin group (0.05 mg/kg), and fluoxetine group (10 mg/kg), with 10 rats in each group, and all of them received intraperitoneal injections for four weeks. Neuropeptide Y (NPY), agouti-related protein (AgRP), cannabinoids receptor 1 (CB1R) were detected in the arcuate nucleus (ARC) of the hypothalamus by Western blot. Immunofluorescence was used to detect the NPY/CB1R and AgRP/CB1R co-expression, and real-time fluorescence quantitative PCR was used to detect the mRNA expression of γ-aminobutyric acid A receptor (GABAAR) and γ-aminobutyric acid B receptor (GABABR). The results showed that compared with the normal group, the protein expression of NPY/AgRP/CB1R were all reduced in the model group (P < 0.01, P < 0.001), the fluorescence co-localization of NPY and AgRP with CB1R was reduced, respectively (P < 0.05, P < 0.001), and GABAAR mRNA expression was reduced in the model group, and the GABABR mRNA expression was elevated (P < 0.05 P < 0.01); compared with the model group, NPY/AgRP/CB1R protein expression was elevated in the administered group (P < 0.01, P < 0.001), NPY and AgRP fluorescence co-localization intensity with CB1R, respectively, was elevated (P < 0.01, P < 0.001), GABAAR mRNA expression was elevated (P < 0.05), and GABABR mRNA expression decreased (P < 0.05, P < 0.01). The above results suggest that capsaicin can improve the depressive behavior and feeding behavior of CUMS depressed rats, and its mechanism may play a regulatory role by up-regulating the expression of NPY/AgRP/CB1R/GABAAR and down-regulating the expression of GABABR receptor in the ARC brain region of CUMS depressed rats.
